The Purinosome: A Case Study for a Mammalian Metabolon

Summary: This article introduces a new mechanism by which cells achieve greater efficiency in de novo purine biosynthesis, called the purinosome. The purinosome is a dynamic complex that compartmentalizes purine biosynthetic enzymes. Tumor Reliance on Cytosolic versus Mitochondrial One-Carbon Flux Depends on Folate Availability

Summary: The study showed that under physiological folate levels in the cell environment, cytosolic serine-hydroxymethyltransferase (SHMT1) is the main source of 1C units in various cancers, while mitochondrial 1C flux is excessively suppressed. Tumor-specific reliance on cytosolic 1C flux is associated with poor capacity to retain intracellular folates, determined by the expression of SLC19A1.

Summary: Our study revealed the importance of different levels and types of folate transporters in determining cellular folate accumulation, with PCFT and RFC prevailing over FR alpha in certain conditions. The interplay among these major folate transport systems, depending on transporter levels and extracellular pH, affects the uptake and anti-tumor efficacies of targeted and untargeted antifolates.
