This review focuses on the therapeutic potential of pyrrole and pyrrolidine, particularly in anticancer, anti-inflammatory, antiviral, and antituberculosis applications. It also summarizes recent reports on pyrrole and pyrrolidine analogs and their biological potential.

Summary: The present study focuses on the synthesis and characterization of a new butylidine linked hetero dimer of pyridazinone (PNP). The crystal structure analysis reveals the presence of 1D linear chains connected by C-H···O interaction and interlayer connectivity supported by the unconventional orthogonal dipolar interaction via nitro-aromatic (ONO2 & ···pi(C)Ar). The influence of crystal structure on the photophysical properties of a solid state emitter containing brick layer packing is discussed, and the intermolecular interactions stabilizing the crystal packing are explored using Hirshfeld surface analysis and two-dimensional fingerprint plots.

Identification of 2-substituted pyrrolo[1,2-b]pyridazine derivatives as new PARP-1 inhibitors

Additionally, the pharmacokinetic properties of compound 15b were also investigated.

The compounds showed minimal phytotoxicity on Triticum rootlets and induced moderate to high toxicity on Daphnia magna, similar to nifedipine. The newly synthesized pyrrole derivatives are promising molecules with biological activity and low acute toxicity.

Summary: The oxidative [3+2]-annulation reactions between nitroalkenes and pyridazinium ylides mediated by Cu(OAc)(2).H2O were explored, leading to functionalized pyrrolo[1,2-b]pyridazines and pyrrolo[1,2-a]phthalazines. Mechanistic features of the reaction were studied to support the stepwise [3+2]-cycloaddition/oxidation/HNO2 elimination mechanism, with partially saturated primary [3+2]-adducts identified as key intermediates.

Anticancer Activity of Some Ruthenium(III) Complexes with Quinolone Antibiotics: In Vitro Cytotoxicity, Cell Cycle Modulation, and Apoptosis-Inducing Properties in LoVo Colon Cancer Cell Line

Summary: Our study focused on the anticancer activity of several ruthenium (Ru) complexes with quinolone antibiotics in colon tumor cell cultures. The results showed dose-dependent increased levels of cell lysis and induction of apoptosis in LoVo cancer cells treated with the Ru(III) complexes. Additionally, a major decrease in cell proliferation, with a G0/G1 cell arrest and modulation of cell cycle phases, was observed, indicating the potential for the Ru(III) complexes in future chemotherapeutic approaches.
