Macrophage: Key player in the pathogenesis of autoimmune diseases

Shuang Yang et al.

Summary: Macrophages are essential for the innate immune system and play a crucial role in being the link between innate and adaptive immune responses. They have various functions such as immune tolerance, fibrosis, inflammatory response, angiogenesis, and phagocytosis of apoptotic cells. The dysfunction of macrophages is a significant cause for the occurrence and development of autoimmune diseases. This review mainly focuses on the functions of macrophages in autoimmune diseases, especially in systemic lupus erythematosus (SLE), rheumatoid arthritis (RA), systemic sclerosis (SSc), and type 1 diabetes (T1D), providing references for the treatment and prevention of autoimmune diseases.

Summary: The GENCODE project annotates human and mouse genes and transcripts with high accuracy, using experimental data and bioinformatic tools, and continues to improve annotation infrastructure and tools for the human and mouse genomes. This includes manual annotation for the mouse reference genome, targeted improvements for SARS-CoV-2 related genes, collaborative projects for reference annotation databases, and the first GENCODE supervised automated annotation of lncRNAs.

Folylpoly-γ-glutamate synthetase association to the cytoskeleton: Implications to folate metabolon compartmentalization

Michal Stark et al.

Summary: The study reveals that cytoplasmic FPGS is associated with the cytoskeleton and unconventional myosin-1c, supporting the dynamic channeling of polyglutamylated folates to metabolon compartments. This novel finding enhances folate-dependent de novo biosynthesis of nucleotides and DNA/protein methylation.

The Macrophage Reprogramming Ability of Antifolates Reveals Soluble CD14 as a Potential Biomarker for Methotrexate Response in Rheumatoid Arthritis

Sara Fuentelsaz-Romero et al.

Summary: The research showed that OCM blockers can alter the function and gene expression profile of human macrophages, reprogramming them into a state of LPS tolerance. In early RA patients, MTX-responsive RA patients exhibit lower sCD14 serum levels, with baseline sCD14 levels predicting MTX response.

Metabolic Control of Autoimmunity and Tissue Inflammation in Rheumatoid Arthritis

Jingtao Qiu et al.

Summary: Rheumatoid arthritis (RA) develops in three distinct stages, involving dysfunction of T cells and B cells, tissue invasion, and tissue destruction. Metabolic mis-regulation is a key pathogenic pathway in all phases of RA, with defective DNA repair and misbalanced metabolic pathways contributing to autoimmunity and tissue inflammation. Targeting metabolic abnormalities may offer a novel treatment strategy for autoimmune diseases like RA.

Computational comparison of common event-based differential splicing tools: practical considerations for laboratory researchers

Ittai B. Muller et al.

Summary: This study compared the computational performance of three splicing tools and found that rMATS outperformed MISO and SUPPA2 in terms of job times and CPU utilization. Despite high concordance between tool outputs, differences were observed in the analysis of retained intron events. Users should consider factors such as job time, replicates, and splice event type when choosing the optimal alternative splicing tool for RNA-seq analyses.

Macrophages: The Good, the Bad, and the Gluttony

Ewan R. A. Ross et al.

Summary: Macrophages play critical roles in sterile inflammation and autoimmune diseases, particularly in the pathophysiology of diseases like rheumatoid arthritis. Imbalance in macrophage homeostasis contributes to chronic inflammation, and therapeutic strategies targeting macrophage plasticity are being explored. Apoptotic cell-derived extracellular vesicles may play a role in regulating macrophage phenotype and are a potential target for new therapeutic approaches.
