Computer-Aided Detection of Quantitative Signatures for Breast Fibroepithelial Tumors Using Label-Free Multi-Photon Imaging

Kana Kobayashi-Taguchi et al.

Summary: This study successfully differentiated fibroepithelial lesions using a combined technique of label-free imaging with multi-photon microscopy and artificial intelligence. Tissue sections were detected using multi-photon excited autofluorescence and second harmonic generation (SHG) signals, and a deep learning framework was used to automatically separate epithelial and stromal regions. This provides quantitative signatures for epithelial and stromal alterations in breast tissues.

Extended depth-of-field light-sheet microscopy improves imaging of large volumes at high numerical aperture

Kevin Keomanee-Dizon et al.

Summary: ExD-SPIM is an improved light-sheet microscopy strategy that solves the limitation of capturing fluorescence signals in large volumes by extending the depth of field (DOF) of high-NA detection objectives to match the thickness of the illumination light sheet. Compared to conventional light sheet imaging, ExD-SPIM improves the signal-to-noise ratio and reduces the rate of photobleaching. In whole-brain activity imaging, ExD-SPIM enhances signal sensitivity and volumetric coverage.

Wide field light-sheet microscopy with lens-axicon controlled two-photon Bessel beam illumination

Sota Takanezawa et al.

Summary: The use of two-photon excitation in light-sheet microscopy is limited by its narrow excitation range, but a new illumination optics proposed in this study aims to generate longer extent Bessel beams to address this limitation. A microscope constructed with this technique allows for long-term time-lapse observations and high-speed recording with similar to 2-3-μm axial resolution, expanding the applications of two-photon excitation in light-sheet microscopy.
