The functional evolution of architecturally different plant geranyl diphosphate synthases from geranylgeranyl diphosphate synthase

Shuyan Song et al.

Summary: Terpenoids are the largest class of plant metabolites with diverse functions. The synthesis of terpenoids involves enzymes called geranylgeranyl diphosphate synthases (GGPPSs) and geranyl diphosphate synthases (GPPSs). Through phylogenetic analysis and experimental characterization, it was found that the expansion and functional divergence of GPPS genes started in nonvascular plants, leading to the evolution of both homomeric and heteromeric GPPSs. Mutagenesis and simulations revealed amino acid residues crucial for the functional divergence of GPPSs and GGPPSs.

Long-chain acyl-CoA synthetases activate fatty acids for lipid synthesis, remodeling and energy production in Chlamydomonas

Fan Bai et al.

Summary: The study unraveled the crucial roles of three Chlamydomonas LACS enzymes in lipid synthesis, remodeling, and catabolism through genetic, biochemical, and physiological analyses. It highlighted the importance of lipid homeostasis in cell growth under nutrient fluctuations, with solid experimental evidence supporting the findings. Transcriptional reprogramming was not the primary factor for the observed lipid phenotypes, but rather a shift in metabolic adjustment was suggested.

The role of key genes in astaxanthin biosynthesis in Phaffia rhodozyma by transcript level and gene knockout

Zhipeng Li et al.

Summary: Phaffia rhodozyma is a potential industrial source of natural astaxanthin. This study analyzed the relationship between two pathways (dicyclic and monocyclic) and astaxanthin production in P. rhodozyma. Knockout of CRTYB gene resulted in undetectable astaxanthin levels, indicating its importance in both pathways. Knockout of CRTS gene promoted the monocyclic pathway and increased astaxanthin production.

The damage and tolerance mechanisms of Phaffia rhodozyma mutant strain MK19 grown at 28 °C

Li-Li Miao et al.

Summary: The inhibition of cell growth in P. rhodozyma at temperatures >21 degrees C is due to blocking of DNA, RNA, fatty acid, and cell wall biosynthesis processes. Through metabolic engineering, enhancing the astaxanthin synthesis pathway can improve the adaptability of P. rhodozyma to high temperatures, ultimately increasing astaxanthin production.

Enhancing carbohydrate repartitioning into lipid and carotenoid by disruption of microalgae starch debranching enzyme

Yuichi Kato et al.

Summary: Kato et al. isolated a mutant Chlamydomonas sp. KOR1 with extensive mutations in the ISA1 gene encoding a starch debranching enzyme, leading to improved biofuel production by accelerating carbohydrate repartitioning into lipid and carotenoid under light/dark conditions. The study identified promising targets for metabolic engineering of enhanced lipid accumulation outdoors, showcasing the impact of disrupting DBE on the reparation process.
