The results show that LINC00659 directly targets and negatively regulates miR-525-5p, while miR-525-5p targets Bax. Upregulation of LINC00659 inhibits endothelial cell proliferation and promotes apoptosis, while silencing LINC00659 increases cell viability and inhibits apoptosis through upregulation of miR-525-5p. Further mechanistic studies reveal that miR-525-5p enhances cell viability and inhibits apoptosis by downregulating Bax expression. LINC00659 plays an important role in deep vein thrombosis through regulating endothelial cell apoptosis via the miR-525-5p/Bax axis.

Tissue Factor Pathway Inhibitors as Potential Targets for Understanding the Pathophysiology of Preeclampsia

Summary: This review summarizes the biological functions of TFPI1 and TFPI2 in preeclamptic patients and discusses future directions in preeclampsia research. TFPI1 is an important physiological inhibitor of the TF-initiated extrinsic coagulation pathway, while TFPI2 inhibits fibrinolysis and exerts antifibrinolytic activity. TFPI1 and TFPI2 may play important roles in regulating the coagulation and fibrinolytic system and trophoblast invasion. Concentrations of TF, TFPI1, and TFPI2 are significantly altered in preeclamptic women compared to normal pregnancies. The TFPI protein family may serve as predictive biomarkers for preeclampsia and guide precision therapy.

The results demonstrated that LINC00659 is upregulated in IVC tissues and isolated EPCs of patients with LEDVT, and its knock-down promotes EPC proliferation, migration, and angiogenesis ability.

Significant role of long non-coding RNA MALAT1 in deep vein thrombosis via the regulation of vascular endothelial cell physiology through the microRNA-383-5p/BCL2L11 axis

Summary: This study found that the long non-coding RNA MALAT1 is positively expressed in deep vein thrombosis (DVT) and regulates the behavior of endothelial progenitor cells. The overexpression of MALAT1 suppresses the viability and induces apoptosis in human vascular endothelial cells (HUVECs), while downregulating Bcl-2 expression and upregulating Bax expression. On the other hand, silencing MALAT1 has the opposite effect. Furthermore, the study reveals the interaction among MALAT1, miR-383-5p, and BCL2L11, and demonstrates that miR-383-5p promotes cell proliferation and reduces cell apoptosis in HUVECs by targeting BCL2L11.

Summary: Long noncoding RNAs (lncRNAs) are extensively transcribed and studied, but those overlapping with mRNA in the sense orientation have been less explored. We investigated the lncRNA DAPALR overlapping with the 5' UTR of the male determining gene Doublesex1 (Dsx1) in Daphnia magna. Through affinity purification, we identified an RNA binding protein, Shep, as a DAPALR binding protein. Shep binds to Dsx1 5' UTR by recognizing the overlapping sequence and suppresses mRNA translation. In vitro and in vivo analyses indicated that DAPALR enhances Dsx1 translation efficiency by sequestering Shep. This mechanism may play a significant role in displaying dimorphic gene expressions, such as sex determination, and could explain binary expression in various developmental processes.
