A newly constructed Agrobacterium-mediated transformation system based on the hisB auxotrophic marker for genetic manipulation in Aspergillus niger

Summary: In this study, an ATMT system based on the histidine auxotrophic mechanism was established for A. niger by deleting the hisB gene. The results showed that the ATMT system could achieve 300 transformants per 10^7 fungal spores under optimal transformation conditions. The system was successfully applied for expressing a fluorescent protein-encoding gene and gene targeting in A. niger.

T- DNA insertion mutagenesis in Penicillium brocae results in identification of an enolase gene mutant impaired in secretion of organic acids and phosphate solubilization

Summary: Phosphate-solubilizing fungus Penicillium brocae P6 was isolated from farmland in Guangdong Province, China. A T-DNA insertion population of approximately 4500 transformants was generated by Agrobacterium tumefaciens-mediated transformation to study the phosphate solubilization mechanisms. A mutant (MT27) impaired in phosphate solubilization was obtained and T-DNA insertion site was identified in an enolase gene. The enolase gene was found to be involved in the production of specific organic acids that act as phosphate solubilizing agents.

Identification and Characterization of Neocosmospora silvicola Causing Canker Disease on Pinus armandii in China

swnk plays an important role in the biosynthesis of swainsonine in Metarhizium anisopliae

Summary: This study aimed to analyze the role of the key enzyme swnk in the biosynthesis pathway of Swainsonine (SW) in locoweeds. LC-MS and RT-qPCR were used to determine the concentration of SW and the expression of swnk in the fermentation of Metarhizium anisopliae. The results showed that M. anisopliae had the highest SW content and swnk expression on the 5th day of fermentation. Gene knockout and gene complementation experiments demonstrated the crucial role of swnk in SW synthesis. RNAi interference of swnk affected the cell wall formation in M. anisopliae.

Mrhst4 gene, coding for NAD+-dependent deacetylase is involved in citrinin production of Monascus ruber

Summary: In this study, the deletion of Mrhst4, a member of NAD(+)-dependent histone deacetylase (HDAC), was performed to investigate its role in regulating Monascus azaphilone pigments (MonAzPs) and mycotoxin production, as well as the developmental process in Monascus ruber. The results showed that the absence of Mrhst4 did not significantly affect reproduction, morphology, or micro-morphology. However, it significantly increased MonAzPs yields and citrinin content. RT-qPCR analysis revealed that the relative expression of citrinin biosynthetic pathway genes was also increased in the Mrhst4-deleted strain. Western blot assay indicated that the deletion of Mrhst4 led to increased acetylation levels of specific histone modifications. Therefore, MrHst4 is an important regulator of secondary metabolism, especially in citrinin production, in Monascus ruber.

Phylogeny of Leptographium qinlingensis cytochrome P450 genes and transcription levels of six CYPs in response to different nutrition media or terpenoids

Summary: This study identified and analyzed cytochrome P450 (CYP) genes in the transcriptome of Leptographium qinlingensis and revealed a putative common ancestor shared with Grosmannia clavigera. The findings suggest similar nutrient acquisition and terpene metabolism between L. qinlingensis and G. clavigera.

Transcriptome analyses of the Chinese white pine beetle-fungal symbiont Leptographium qinlingensis under terpene stress or growth on host pine sawdust

Summary: This study reveals the tolerance of Leptographium qinlingensis to monoterpenes and diterpenes and its ability to utilize carbon sources in host pine sawdust. The transcriptome analysis identified key genes and enzymes involved in terpene tolerance and nutrition utilization. These findings are important for further understanding the pathogenicity of L. qinlingensis and the molecular interactions between beetle-symbiotic fungi and their host trees.

Diversity of Ophiostomatoid Fungi Associated with Dendroctonus armandi Infesting Pinus armandii in Western China

Summary: The study aimed to identify the diversity of ophiostomatoid fungi associated with D. armandi infesting P. armandii in western China. Seven species belonging to five genera were identified, including three known species and two novel taxa. Ophiostoma shennongense was the dominant taxon. This study provides a valuable scientific theoretical basis for the occurrence and management of D. armandi in the future.

Development of a new Agrobacterium-mediated transformation system based on a dual auxotrophic approach in the filamentous fungus Aspergillus oryzae

Summary: This study successfully constructed a new ATMT system in Aspergillus oryzae by deleting the hisB gene, making genetic transformation more efficient. The dual auxotrophic ATMT system demonstrated high transformation efficiency and enabled the co-expression of multiple selectable markers. This system provides a promising genetic tool for studying recombinant expression and gene function in the industrially important fungus A. oryzae.

Nutrient-Poor Breeding Substrates of Ambrosia Beetles Are Enriched With Biologically Important Elements

Summary: Fungus-farming within galleries in the xylem of trees has independently evolved in multiple lineages of beetles. These ambrosia beetles actively cultivate ambrosia fungi as their main food source, with fungi providing essential nutrients that are lacking in the nutrient-poor environment of the xylem. The study found that the galleries of ambrosia beetles are enriched with rare elements that are scarce in the surrounding xylem, suggesting translocation of nutrients by the fungal associates.
