Pathogenicity and virulence of Entamoeba histolytica, the agent of amoebiasis

Summary: The amoeba parasite Entamoeba histolytica is a widespread protozoan that causes a disease called amebiasis in humans. It has evolved various mechanisms to evade immune responses, interact with microbiota, and utilize host resources for its survival and reproduction. In some cases, the parasite becomes pathogenic and causes invasive infection, facing challenges such as crossing mucus, epithelium, and connective tissue.

Identification and Molecular Characterization of Four New Blastocystis Subtypes Designated ST35-ST38

Summary: Three recent studies found four novel sequences in mammalian hosts infected with Blastocystis, with B. lapemi as the most similar sequence. However, full-length ssu rRNA gene sequences were not available to confirm the validity of these new subtypes. This study used Nanopore MinION sequencing to obtain full-length reference sequences for each of the new subtypes, and confirmed their validity through phylogenetic analysis and pairwise distance comparisons. The proposed subtype designations for the novel sequences in this study are ST35-ST38, which will aid in future accurate sequence descriptions and studies of Blastocystis epidemiology and subtype diversity.

Summary: This study reveals a novel mechanism by which Giardia cysteine proteases modulate goblet cell activity through cleavage and activation of protease-activated receptor 2. Giardia duodenalis (assemblage A) increases MUC2 mucin gene expression in human colonic epithelial cells dependent on protease-activated receptor 2 activation and Giardia cysteine protease activity. Protease-activated receptor 2 can regulate mucin gene expression in intestinal goblet cells.

Immunomodulatory effect of extracellular vesicles from Entamoeba histolytica trophozoites: Regulation of NETs and respiratory burst during confrontation with human neutrophils

Summary: This study isolated and characterized extracellular vesicles (EVs) released by the human protozoan parasite Entamoeba histolytica. The EVs were found to modulate the immune response of neutrophils, reducing oxidative burst and NET release. Proteomic analysis showed differences in protein expression and abundance between EVs from amoebae and neutrophils. These findings highlight the importance of EVs in the immunomodulatory effects exerted by amoeba on neutrophils.

Association of Blastocystis ST6 with higher protease activity among symptomatic subjects

Summary: The study indicated a higher protease activity among isolates from symptomatic subjects, suggesting a functional role for proteases in clinical symptoms. However, there was no significant correlation between protease activity of different subtypes and pro-inflammatory biomarker expression levels. The expression patterns of pro-inflammatory biomarkers in cell line also varied based on the presence of symptoms and time points.

Advances in transcriptomic and proteomic technologies have allowed for the assembly of draft genome sequences for different assemblages of G. duodenalis, leading to the identification of key virulence markers. Some drugs and vaccines targeting virulence factors have been developed, offering potential alternatives to current treatment options against giardiasis.
