DNA barcoding of Cymbidium by genome skimming: Call for next-generation nuclear barcodes

Le Zhang et al.

Summary: In this study, DNA barcoding was conducted for species identification of Cymbidium using plastomes. A comprehensive DNA barcode reference library for Cymbidium species identification was established. The plastome showed improved species identification rates compared to standard DNA barcodes, but still had limitations. Nuclear genome data were also explored for species identification and showed potential as next-generation nuclear barcodes.

Comparative Analyses of 35 Complete Chloroplast Genomes from the Genus Dalbergia (Fabaceae) and the Identification of DNA Barcodes for Tracking Illegal Logging and Counterfeit Rosewood

Zhou Hong et al.

Summary: The study focuses on the identification and tracking of rosewood using molecular methods due to the rarity of these trees in the wild and the growing international illicit trade. Through sequencing and comparative analysis of chloroplast genomes, unique genetic markers were identified for Dalbergia species. Phylogenetic analysis supported the classification of certain species based on morphological traits. The study also highlighted the discrepancies between taxonomic determinations and phylogenetic tree topology.

Comparative phylogenetic analysis of complete plastid genomes of Renanthera (Orchidaceae)

Tao Xiao et al.

Summary: This study reports on the de novo assembled and annotated plastome of four species of Renanthera, a genus of orchids with significant ornamental value. The plastomes showed a typical quadripartite structure and nucleotide variations that could distinguish the species. Phylogenetic analyses revealed the relationship between Renanthera and other related genera. The genetic data obtained from this study can contribute to species identification and further research on the phylogenomics of Orchidaceae.

On the Edge of Dispensability, the Chloroplast ndh Genes

Bartolome Sabater

Summary: The absence of ndh genes in many species of eukaryotic algae and certain vascular plants provides insights into the natural selection of photosynthesis-related genes in evolution. Plants without ndh genes are on the verge of extinction due to variable evolutionary environments, and most extant species may have recently lost these genes. Additional DNA sequencing and investigations into molecular mechanisms in Gymnosperms lacking ndh genes will help develop a unified model of the evolutionary and functional consequences of their absence.

Chloroplast Genomes of Two Species of Cypripedium: Expanded Genome Size and Proliferation of AT-Biased Repeat Sequences

Yan-Yan Guo et al.

Summary: This study presented the sequencing, assembly and annotation of the chloroplast genomes of Cypripedium tibeticum and Cypripedium subtropicum, revealing their expanded genome size, low GC content, proliferation of AT-rich repeat sequences, and gene density. The increase in genome size is attributed to the proliferation of AT-biased non-coding regions, indicating a typical example of plastome expansion induced by non-coding regions in the genus. Hybrid assembly based on long and short reads is recommended for sequencing plastomes with AT-biased base composition.

Comparative and Phylogenetic Analysis of the Complete Chloroplast Genome of Santalum (Santalaceae)

Xiaojin Liu et al.

Summary: Santalum, a hemiparasitic genus known for its aromatic heartwood oil, is facing threats due to over-harvesting for sandalwood-based products. A study sequencing the chloroplast genomes of 11 Santalum species revealed high similarity among them, with the genome containing 110 unique genes, some of which have lost their functions. Divergence time analysis suggests that Santalum species likely originated through radiation evolution, with most speciation events occurring less than 1 Mya.
