Structural determinants of the interactions of catechins with Aβ oligomers and lipid membranes

Rashik Ahmed et al.

Summary: Catechins can reduce the toxicity of intrinsically disordered protein oligomers by modulating their interactions with membranes. The study reveals multiple drivers of catechin affinity for Alzheimer's-associated amyloid-beta oligomers and model membranes, and suggests that covalent changes can lead to unexpected increases in affinity.

A Small Molecule Stabilizes the Disordered Native State of the Alzheimer's Aβ Peptide

Thomas Lohr et al.

Summary: The stabilization of native states of proteins is a powerful drug discovery strategy. This study reveals that a small molecule can stabilize the intrinsically disordered state of the A beta 42 peptide associated with Alzheimer's disease through a disordered binding mechanism. This mechanism involves both enthalpic gain from local interactions and entropic gain from global effects.

AlphaFold2: A Role for Disordered Protein/Region Prediction?

Carter J. Wilson et al.

Summary: The development of AlphaFold2 has brought a paradigm shift in structural biology and has shown excellent performance in predicting disordered regions. The accuracy of the constructed disorder predictor from AlphaFold2 structures varies, with the naive assumption of assigning residues to ordered or disordered based on helices, strands, and H-bond stabilized turns leading to a significant overestimation of disorder. The predicted local distance difference test (pLDDT) provides a reliable measure of residue-wise disorder and shows an interesting relationship with secondary structure.

Determination of Anti-Alzheimer's Disease Activity of Selected Plant Ingredients

Tomasz Tuzimski et al.

Summary: Neurodegenerative diseases, including Alzheimer's disease, pose a significant public health challenge due to the aging population. Discovering new treatment options is crucial, and controlling acetylcholine levels using acetylcholinesterase inhibitors has shown promise. However, these drugs have side effects, driving the need for new, more effective drugs. Plant-derived compounds offer a potential solution, with lower toxicity compared to synthetic agents. This review aims to identify safe and effective compounds for further testing to develop better treatments for Alzheimer's disease.

Constructing conformational library for amyloid-β42 dimers as the smallest toxic oligomers using two CHARMM force fields

Maryam Haji Dehabadi et al.

Summary: The study of Amyloid-beta (A beta) dimers in Alzheimer's disease is of great interest. The low stability of these dimers and their tendency to aggregate have limited the structural characterization. In this study, a computational protocol called blockwise excursion sampling (BES) was used to efficiently generate a conformational library of A beta(42) dimers, which is important for finding potential drug candidates.

Summary: This study utilizes an A beta dimer library and molecular dynamics simulations to investigate the interactions between curcumin degradation products and A beta dimer. The simulations reveal that aromatic residues of A beta, particularly (FF20)-F-19, interact with ferulic aldehyde and vanillin through pi-pi stacking, resulting in significant structural changes in the (KLVFF20)-K-16 region.

Characterization of the Conformations of Amyloid Beta 42 in Solution That May Mediate Its Initial Hydrophobic Aggregation

Krushna Sonar et al.

Summary: Intrinsically disordered peptide A beta 42 is associated with formation of amyloid plaques and Alzheimer's disease. The mechanism of oligomerization of A beta 42 is not well understood due to its structural disorder. Molecular dynamics simulations reveal that A beta 42 conformations with alpha-helical structure have a higher probability of initiating aggregation, shedding light on the mechanism of oligomerization of this disordered peptide.

Insights into Molecular Mechanisms of EGCG and Apigenin on Disrupting Amyloid-Beta Protofibrils Based on Molecular Dynamics Simulations

Mei Fang et al.

Summary: This study investigates the molecular mechanisms of (-)-epigallocatechin-3-gallate (EGCG) and apigenin in disrupting Afi42 protofibrils using molecular dynamics simulations. Both EGCG and apigenin can decrease the proportion of beta-sheet and bend structures and induce random coil structures in Afi42 protofibrils. Through disrupting hydrogen bonds, salt bridges, van der Waals interactions, and electrostatic interactions, EGCG and apigenin have the ability to destabilize Afi42 protofibrils.
