Enhanced anticancer activity of encapsulated geraniol into biocompatible lipid nanoparticles against A549 human lung cancer cells

Summary: Geraniol (GOH), a monoterpene alcohol found in essential oils, was encapsulated into nanostructured lipid carriers (NLC) to improve its anticancer activity. The NLC/GOH exhibited high stability and low dispersity, and showed increased release of GOH at acidic pH. The NLC/GOH were safe, absorbed serum proteins, and preferentially interacted with human serum albumin. A time-dependent cellular uptake of NLC/GOH was observed in A549 cells, leading to increased cytotoxicity, mitochondrial membrane depolarization, and cell death. NLC/GOH showed highly cytotoxic concentrations against A549 cells but was non-toxic to normal lung fibroblasts. Additionally, GOH nanoencapsulation into NLC potentiated the inhibition of A549 cell migration.

Incorporation of glycyrrhizic acid and polyene phosphatidylcholine in lipid nanoparticles ameliorates acute liver injury via delivering p65 siRNA

Summary: In this study, a new strategy of incorporating glycyrrhizic acid (GA) and polyene phosphatidylcholine (PPC) into lipid nanoparticles (GA/PPC-modified LNPs) was developed, which showed enhanced cellular uptake, improved gene-silencing efficiency, reduced cytotoxicity, and increased siRNA stability. GA/PPC-modified LNPs targeting NF-kappa B attenuated acute liver injury and facilitated intracellular delivery of antisense oligonucleotides (ASOs) and mRNA inhibiting viral infection. These findings suggest that GA/PPC-modified LNPs could serve as a promising nucleic acid delivery system.

The optimized formulation exhibited desirable aerodynamic properties and inhibited TGF-beta signaling, indicating its potential as a treatment for PAH.

Summary: In this study, a novel corn oil-based nanoemulsion encapsulated with Yellow Monascus Pigment crude extract (CO-YMPN) was successfully prepared. The optimized fabrication conditions were a emulsifier ratio of 5:3 (Tween 60 to Tween 80) and a Yellow Monascus Pigment crude extract (YMPCE) concentration of 20.00% wt%. CO-YMPN showed excellent DPPH radical scavenging capability and improved lipase hydrolysis capacity. It also exhibited excellent storage stability and water solubility in the final water system.

Caffeic acid loaded into engineered lipid nanoparticles for Alzheimer's disease therapy

Summary: Alzheimer's disease (AD) is an incurable neurological illness and the leading cause of dementia, characterized by amyloid beta (Aβ) fibril deposits. Caffeic acid (CA) has potential value for AD therapy due to its antiamyloidogenic, anti-inflammatory, and antioxidant properties. Liposomes loading CA were produced and Tf was conjugated to the liposomes' surface to direct the nanoparticles (NPs) to the blood-brain barrier. The optimized Tf-modified NPs exhibited suitable physical properties and encapsulation efficiency, and showed sustained release of CA for 8 days. The drug delivery system (DDS) can prevent Aβ aggregation and fibril formation, and disaggregate mature fibrils, indicating its potential for preventing and treating AD.

Chondroitin/Lactoferrin-dual functionalized pterostilbene-solid lipid nanoparticles as targeted breast cancer therapy

Summary: Breast cancer, the leading cause of cancer-associated mortality in women, requires novel therapeutic approaches. Pterostilbene (PTS), a phytochemical agent, shows promise against breast cancer but has limitations. In this study, PTS-loaded solid lipid nanoparticles (PTS-SLNs) were developed with active-targeting using lactoferrin (Lf) and chondroitinsulfate (CS). CS/Lf/PTS-SLNs exhibited desirable properties and demonstrated superior cytotoxicity, anti-migratory effect, and cellular uptake compared to PTS-solution in vitro. In an orthotopic cancer model, CS/Lf/PTS-SLNs showed improved anti-tumor efficacy and molecular-level effects. These findings highlight CS/Lf/PTS-SLNs as a promising nanoplatform for targeted-breast cancer therapy.

Assessment of anti-psoriatic activity of bakuchiol-loaded solid lipid nanoparticles-based gel: design, characterization, and mechanistic insight via NF-kB signaling pathway

Summary: The aim of this study was to evaluate the anti-psoriatic potential of bakuchiol loaded solid lipid nanoparticles (SLNs) by modulating inflammatory and oxidative pathways. Bak-loaded SLNs were prepared and characterized. The release studies confirmed sustained release of Bak-SLNs-based gel. In a UV-B-induced psoriatic rat model, Bak showed significant anti-psoriatic effect via regulating inflammatory markers and levels of antioxidant enzymes. RT-qPCR analysis confirmed the downregulation of inflammatory markers by Bak. The study indicates that Bak-loaded SLNs-based gel can be a novel therapeutic approach for psoriasis.

Umbelliferone loaded PEGylated liposomes: preparation, characterization and its mitigatory effects on Dalton's ascites lymphoma development

Summary: The aim of this study was to investigate the capability of MGN-NLC as a potential drug delivery system for ocular administration. The in vitro and ex vivo results showed that MGN-NLC exhibited no cytotoxic effects, maintained the antioxidant role of MGN, and could permeate and accumulate in ocular tissues. These findings suggest a potential application of MGN-NLC in the treatment of oxidative stress-related ocular diseases.

Encapsulation of sesamol and retinol using alginate and chitosan-coated W/O/W multiple emulsions containing Tween 80 and Span 80

Summary: The production conditions of multiple W/O/W nanoemulsions containing sesamol and retinol were optimized using response surface methodology (RSM). Span 80, Tween 80, and water in oil ratio (W/O) were considered as independent variables, while encapsulation efficiency (EE%) and particle size were taken as dependent variables. An optimum sample with high EE% and small particle size was produced by adjusting the percentages of Span 80, Tween 80, and W/O. The release of sesamol and retinol was measured in simulated gastric fluid (SGF) and simulated intestinal fluid (SIF), showing different release rates.

Development of thymol nanoemulsions with potential application in oral infections
