Acute ATP loss during irreversible electroporation mediates caspase independent cell death

Summary: Irreversible electroporation (IRE) can cause apoptosis, necrosis, oncosis, or pyroptosis. The mode of cell death following IRE is determined by intracellular ATP levels. Cell death after IRE is insensitive to caspase inhibition and is correlated with ATP loss, which cannot be rescued by ATP supplementation.

A perspective on modeling pore energy and pulsed electromagnetic field induced cell membrane perforation

Summary: This perspective paper reviews the theoretical models of reversible and irreversible electroporation induced by microsecond and high-frequency nanosecond pulsed electric fields, as well as the theoretical model of pulsed electric field electroporation considering the mechanical properties of cell membranes. Additionally, it proposes a theoretical model of cell membrane magnetoporation under the action of a pulsed magnetic field, which provides a theoretical foundation for the popularization and application of non-contact cell membrane perforation technology.

Summary: This study observed the effects of pulsed 3.5 GHz high-power microwave on normal brain astrocytes and cancer cells, and investigated the underlying mechanisms. The results showed that high-power microwave radiation produced reactive oxygen species, affecting cell viability, mitochondrial activity, and cell death rates. 60 pulses were found to be harmful, while 25 pulses showed therapeutic effects on cancer cells.

However, recent studies have shown that pulse duration and type can be more flexible in achieving electrochemotherapy (ECT), gene electrotransfer (GET), and irreversible electroporation (IRE). The activation of the immune system in these therapies can be controlled through pulse duration, as different pulse durations and types can result in different DAMP release and immune responses.

High-Intensity Pulsed Electromagnetic Field-Mediated Gene Electrotransfection In Vitro

Summary: This study demonstrates the feasibility of using high-intensity pulsed electromagnetic field (HI-PEMF) for non-viral delivery of genetic material in vitro. The results show that HI-PEMF can cause DNA adsorption to the cell membrane and induce gene electrotransfection. Endocytosis is also identified as a possible mechanism for introducing large molecules into cells by HI-PEMF.

Contactless delivery of plasmid encoding EGFP in vivo by high-intensity pulsed electromagnetic field

Summary: The study demonstrates the feasibility of using HI-PEMF treatment as a noninvasive approach to achieve in vivo transfection by transporting larger molecules such as pDNA encoding EGFP into muscle and skin. Long-term expression of EGFP in the muscle and skin was achieved after HI-PEMF treatment, with significantly less infiltration of inflammatory mononuclear cells in muscle tissue compared to conventional gene electrotransfer. The antitumor effectiveness of HI-PEMF for electrotransfer of therapeutic plasmid was also demonstrated, showing great potential in gene therapy and DNA vaccination.

Summary: The study found that pulsed high magnetic field stimulation can increase the reversible permeability of the BBB, with a more significant effect seen with 28 pulses at 1 Hz frequency. Changes in EBD uptake and MRI signal intensity in the exposed side peaked within 0.5-1 hour after exposure to the effective PHMF stimulation protocol (28 pulses at 1 Hz frequency) and returned to normal levels within 24 hours.
