Metabolic Fingerprints of Effective Fluoxetine Treatment in the Prefrontal Cortex of Chronically Socially Isolated Rats: Marker Candidates and Predictive Metabolites

Chronic fluoxetine treatment in socially-isolated rats modulates the prefrontal cortex synaptoproteome

Summary: Chronic social isolation (CSIS) and synapse dysfunction are related to the development of major depressive disorder (MDD). Fluoxetine (Flx) is commonly used to treat MDD, but its mechanisms of action are still unclear. This study investigated the changes in protein levels and molecular signaling pathways in the prefrontal cortex of adult male Wistar rats exposed to CSIS and treated with Flx using synaptoproteomics. The findings showed that Flx treatment decreased levels of proteins involved in vesicle-mediated transport and increased levels of exocytosis-associated proteins, while CSIS reduced levels of proteins involved in ATP metabolism, endocytosis, and proteolysis. Flx treatment in CSIS rats increased the regulation of proteins involved in synaptic plasticity, mitochondrial functionality, and ion transport, alleviating CSIS-induced impairment.
