Multistability in Macrophage Activation Pathways and Metabolic Implications

Carsten Geiss et al.

Summary: Macrophages have reversible activation states called M1 and M2, which are important for understanding inflammatory diseases and therapies. This review explores evidence for bistability in macrophage activation pathways and suggests targeting metabolic environments as a potential therapeutic approach for modulating macrophage transitions.

Protective role of Cytoglobin and Neuroglobin against the Lipopolysaccharide (LPS)-induced inflammation in Leydig cells ex vivo

Derya Sagrac et al.

Summary: CYGB and NGB proteins play an important role in protecting Leydig cells from dysfunction under inflammatory and oxidative stress conditions. Downregulation of CYGB and NGB leads to increased release of nitride oxide, blocked cell cycle progression, reduced testosterone production, and increased expression of inflammatory and apoptotic pathway genes. On the other hand, overexpression of CYGB and NGB reduces the expression of inflammatory proteins, increases the expression of testosterone biogenesis pathway genes, and improves testosterone production.

A label-free SPR biosensor for specific detection of TLR4 expression; introducing of 10-HDA as an antagonist

Farshid Eslami-Kaliji et al.

Summary: In this study, a new configuration of surface plasmon resonance (SPR) biosensor was successfully used to detect the interaction between Toll-like receptor 4 (TLR4) and 10-hydroxy-2-decanoic acid (10-HDA). The SPR angle was significantly reduced in the presence of TLR4-expressing cells, indicating the specific interaction between TLR4 and 10-HDA. This research provides important evidence for the development of antagonists to inhibit TLR4-induced immune cell activation.

Synergistic anti-inflammatory effects of graphene oxide quantum dots and trans-10-hydroxy-2-decenoic acid on LPS-stimulated RAW 264.7 macrophage cells

Minjie Huang et al.

Summary: This study investigated the effects of graphene oxide quantum dots (GOQDs) on the immune function of macrophages and found that they promoted the anti-inflammatory effect of 10-HDA and affected amino acid metabolism and lipid metabolism in these cells. These findings provide a basis for the further application of GOQDs in biomedical fields.

Metabolic reprogramming in macrophage responses

Yang Liu et al.

Summary: Macrophages are key regulators of tissue homeostasis, involved in tissue development, repair, and defense against pathogens. TAMs play a crucial role in the tumor microenvironment, impacting tumor initiation, growth, invasion, and metastasis. The metabolic characteristics of macrophages influence their phenotype and function, with pro-inflammatory M1 macrophages relying on glycolysis and anti-inflammatory M2 macrophages more dependent on mitochondrial OXPHOS.

Anti-Tumor Effects of Queen Bee Acid (10-Hydroxy-2-Decenoic Acid) Alone and in Combination with Cyclophosphamide and Its Cellular Mechanisms against Ehrlich Solid Tumor in Mice

Aishah E. Albalawi et al.

Summary: The study demonstrated that treatment with 10-HDA at doses of 2.5 and 5 mg/kg, especially in combination with CP, significantly reduced tumor growth, tumor markers, serum liver and kidney levels, lipid peroxidation and nitric oxide, TNF-alpha levels, and Bcl-2 expression. Additionally, 10-HDA in combination with CP also significantly improved antioxidant enzyme levels and expression of caspase-3 and Bax genes.
