The Role of CXC Chemokines in Cancer Progression

Tiantian Wu et al.

Summary: CXC chemokines are small chemotactic and secreted cytokines that are dysregulated in multiple types of cancer and closely correlated with tumor progression. They play crucial roles in promoting angiogenesis, stimulating inflammatory responses, and facilitating tumor metastases. They have the potential to serve as diagnostic and prognostic biomarkers, as well as therapeutic targets for cancer treatment.

CD38: A Significant Regulator of Macrophage Function

Wentao Li et al.

Summary: CD38 is a cell surface glycoprotein and extracellular enzyme that plays a crucial role in immune system regulation. It acts as an NADase, producing adenosine for immunosuppression, and is required for immune cell activation and proliferation. CD38 also affects the aggregation and polarization of macrophages, and is involved in the regulation of macrophage function.

Recombinant Viral Vectors for Therapeutic Programming of Tumour Microenvironment: Advantages and Limitations

Karina Spunde et al.

Summary: Viral vectors are being studied as tools for cancer immunotherapy, but their clinical success is currently limited. Understanding the tumor immune state and developing vectors to modify the immunosuppressive tumor microenvironment are key elements for successful virus-based cancer immunotherapy. This review discusses DNA and RNA virus vectors delivering immunomodulatory genes and their potential to switch the tumor microenvironment to an immune-responsive state. The advantages and limitations of virus vectors for targeted therapeutic programming of tumor immune cell populations and tumor stroma are highlighted.

Targeting MARCO and IL37R on Immunosuppressive Macrophages in Lung Cancer Blocks Regulatory T Cells and Supports Cytotoxic Lymphocyte Function

Linnea La Fleur et al.

Summary: The research revealed the correlation between MARCO-expressing TAMs and poor clinical outcomes in patients with non-small cell lung cancer. These TAMs block the antitumor activities of NK cells and T cells, and gain an immune-suppressive phenotype through the release of IL37. Targeting MARCO or IL37R may remodel the immune-suppressive microenvironment in lung cancer patients.

Lipopolysaccharide promotes metastasis via acceleration of glycolysis by the nuclear factor-κB/snail/hexokinase3 signaling axis in colorectal cancer

Xuesong Wu et al.

Summary: LPS activates inflammasomes in cancer cells through NF-kappa B and promotes metastasis through enhanced glycolysis by the NF-kappa B/Snail/HK3 signaling pathway in CRC. Metformin could prevent this effect of LPS.

Immune cell and tumor cell-derived CXCL10 is indicative of immunotherapy response in metastatic melanoma

Robin Reschke et al.

Summary: In human melanoma metastases, CD45(+) cells and Sox10(+) cells are identified as the major cellular sources for CXCL10. CXCL10 within the tumor site serves as a positive prognostic factor for response to immunotherapy, and the use of RNAscope technique is feasible in paraffin tissue. Strategies supporting effector T cell recruitment through the induction of CXCL10 may enhance the efficacy of immunotherapy.
