Eda controls the size of the enamel knot during incisor development

Summary: This study investigates the formation of incisor germs in Eda-/- mice through 3D reconstruction, expression analysis, cell lineage tracing experiments, and western blot analysis. The results show that the absence of functional Eda protein has minimal impact on the early expression of Shh in the incisor, but affects the later expression of Shh in the primary enamel knot of the functional tooth. Eda deficiency leads to a smaller region of Shh expression and reduced contribution from Shh descendant cells, resulting in the formation of an abnormal enamel organ and a hypoplastic functional incisor. Eda appears to influence the spatial formation of successional signalling centers during odontogenesis.

PAX9 mutations and genetic synergism in familial tooth agenesis

Loss-of-function mutations in PAX9 and WNT10A cause FTA, and the presence of both mutations leads to more severe phenotypes due to mutational synergism. This study identified new disease-causing mutations in PAX9 and expanded the knowledge of phenotypic and genotypic spectrums of PAX9-associated disorders, providing insights into the molecular mechanism of FTA variable expressivity.

Fibulin-1 Regulates Initiation of Successional Dental Lamina

Summary: The successional dental lamina (SDL) of permanent teeth is maintained in a quiescent state until adolescence. Biomechanical stress generated by the rapid growth of deciduous teeth inhibits SDL development. A specific fibroblast subtype, DFDP, in the dental follicle mesenchyme was identified, which is related to biomechanical stress and plays a role in maintaining SDL niche homeostasis. The biomechanical stress-mediated RUNX2-fibulin-1 axis inhibits SDL initiation.

MSX1 Drives Tooth Morphogenesis Through Controlling Wnt Signaling Activity

Summary: Tooth agenesis is a common birth defect that occurs due to morphogenesis failure in early tooth development. This study shows that Msx1 regulates Wnt signaling by suppressing the expression of Dkk2 and Sfrp2, and that Sostdc1/Wise also plays a role in the odontogenic pathway downstream of Msx1. The findings suggest that modulation of Wnt signaling by Dkk2 and Sostdc1 is crucial for proper tooth morphogenesis.

Summary: Tooth development in mammals is regulated by the enamel knot signalling center. While there are differences in signalling, the presence of an enamel knot in non-mammalian vertebrates is still debated. Through studying sharks, researchers found similarities in the expression of fgf3, fgf10, and shh, as well as changes in tooth phenotypes following perturbations of canonical Wnt signalling, which supports the conservation of the enamel knot-like signalling center throughout vertebrates. This suggests that tooth types in different animals follow a similar developmental plan, and the lineage-specific differences in signalling do not refute the homology of this signalling center.

The regulation of Msx1 by BMP4/pSmad1/5 signaling is mediated by importin7 in dental mesenchymal cells

Summary: The study found that IPO7 is strongly expressed in mouse dental mesenchymal cells at postnatal day 1 (PN1) both in vitro and in vivo. Upon BMP4 stimulation, IPO7 translocated from the cytoplasm to the nucleus. Knockdown of IPO7 inhibited the nuclear accumulation of pSmad1/5 in response to BMP4 stimulation.
