These findings provide insights into how DNAJC5 dysfunction leads to lipofuscinosis and neurodegeneration.

Summary: Ferroptosis, a non-apoptotic cell death modality, holds great potential as a strategy to combat therapy-resistant cancers. Cells that develop resistance to conventional therapies or metastatic cancer cells have shown increased sensitivity to ferroptosis. Therefore, studying the regulatory networks of ferroptosis in cancer and utilizing ferroptosis to enhance cancer cell sensitivity offer novel therapeutic opportunities.

PTEN is both an activator and a substrate of chaperone-mediated autophagy

Summary: PTEN is a lipid phosphatase that plays a crucial role in cancer and aging. It regulates chaperone-mediated autophagy (CMA) and is a CMA substrate. Metabolic changes downstream of PTEN require CMA activity. Overexpression of PTEN enhances CMA, while PTEN knockdown reduces CMA. PTEN and CMA are both negative regulators of glycolysis and lipid droplet formation. The protein levels of PTEN are sensitive to CMA and accumulate in lysosomes with elevated CMA. CMA is both an effector and a regulator of PTEN.

LAMP2A, LAMP2B and LAMP2C: similar structures, divergent roles

Summary: LAMP2 is a major protein component of the lysosomal membrane and has three isoforms, LAMP2A, LAMP2B, and LAMP2C, with different distribution and functions. LAMP2A acts as a receptor and channel for transporting cytosolic proteins in chaperone-mediated autophagy. LAMP2B is involved in autophagosome-lysosome fusion and is a component of exosome membranes. LAMP2C participates in a novel type of autophagy that degrades nucleic acids in lysosomes. This review provides a comprehensive summary of the functions and mechanisms of each LAMP2 isoform in various pathophysiological processes and human diseases. The evolutionary patterns of these isoforms in vertebrates are discussed, along with technical guidance for investigating them. Unanswered questions in this research area are also highlighted. Advances in understanding the functions of the three LAMP2 isoforms will reveal new connections between lysosomal dysfunction, autophagy, and human diseases.

Furthermore, CCT2 specifically promotes the autophagic degradation of solid protein aggregates, independent of other ubiquitin-binding receptors or chaperone-mediated autophagy.

In this study, a novel C128Y mutation in the DNAJC5 gene was identified in a young Chinese female with adult-onset NCL, leading to abnormal palmitoylation and lipofuscin deposits.
