Inhibitors of the Ubiquitin-Mediated Signaling Pathway Exhibit Broad-Spectrum Antiviral Activities against New World Alphaviruses

Summary: There are currently no approved therapeutics or vaccines for the treatment or prevention of Venezuelan Equine Encephalitis Virus (VEEV) and Eastern Equine Encephalitis Virus (EEEV) infections. However, inhibitors targeting the ubiquitin proteasome system (UPS) signaling pathways demonstrate broad-spectrum antiviral activity against these viruses. Among the tested inhibitors, NSC697923, bardoxolone methyl, and omaveloxolone exhibit significant inhibitory effects on VEEV and EEEV, suggesting their potential as therapeutic candidates for alphavirus infections.

Characterization of a small molecule inhibitor of disulfide reductases that induces oxidative stress and lethality in lung cancer cells

Summary: Phenotype-based screening led to the identification of a compound called LCS3 that selectively impairs the growth of LUAD cells, induces oxidative stress, and inhibits the activity of disulfide reductases GSR and TXNRD1. The study also reveals that LCS3-sensitive LUAD cells are sensitive to the synergistic inhibition of glutathione and thioredoxin pathways. Furthermore, NQO1 loss is identified as a mechanism of LCS3 resistance through a genome-wide CRISPR knockout screen.
