Targeting Prostate Cancer, the 'Tousled Way'

CRISPR screens reveal genetic determinants of PARP inhibitor sensitivity and resistance in prostate cancer

Summary: Prostate cancer with BRCA1/2 mutations is sensitive to PARP inhibitors, but other DNA damage response gene alterations are not consistently predictive. In this study, CRISPR-Cas9 screens in BRCA1/2-proficient prostate cancer cells identify MMS22L deletion as a biomarker for PARP inhibitor hypersensitivity. Loss of CHEK2, unexpectedly, confers resistance to PARP inhibition through increased expression of BRCA2, but combination treatment with ATR inhibitor overcomes this resistance. These findings have implications for the use of PARP inhibitors and biomarkers in prostate cancer.

Summary: TLK2 selectively targets ASF1 by mimicking its client protein histone H3, promoting nucleosome assembly. This selectivity is achieved through recognition of phosphorylation site motifs in ASF1 by the catalytic domain of TLK2 and interaction between a short sequence at the N-terminus of TLK2 and the N-terminal domain of ASF1.

Summary: The PROfound trial found that treatment with the PARP inhibitor olaparib benefits men with metastatic castration-resistant prostate cancer harboring alterations in 15 different DNA repair genes. However, the benefit of olaparib was less significant, if present at all, for men with prostate cancer harboring non-BRCA DNA repair alterations compared to those with BRCA alterations.

Summary: This study found that enzalutamide combined with androgen deprivation therapy was more effective than bicalutamide for Black men with metastatic hormone-sensitive prostate cancer (mHSPC) in terms of the prostate-specific antigen response rate. The inclusion of enzalutamide in the treatment plan for mHSPC should be strongly considered based on these improved outcomes.

Inactivation of TLK1 leads to reduced AKT phosphorylation, which can be potentiated with AKTIP knockdown. The TLK1 inhibitor J54 inhibits LNCaP cell growth by reducing AKT activation.
