Perfluoroalkyl and Polyfluoroalkyl Substances in Groundwater Used as a Source of Drinking Water in the Eastern United States

Peter B. McMahon et al.

Summary: This study evaluated the occurrence of perfluoroalkyl and polyfluoroalkyl substances (PFAS) in groundwater used as a source of drinking water in the eastern United States. The results showed that 14 out of 24 PFAS were detected in the groundwater samples, with at least one PFAS detection found in 60% of public-supply wells and 20% of domestic wells. The study also identified several factors that were significantly associated with PFAS detections, such as tritium concentration, urban land use, and VOC and pharmaceutical concentrations.

Perfluoroalkyl substances and lipid concentrations in the blood: A systematic review of epidemiological studies

Soon Hoe Ho et al.

Summary: This systematic review examines the associations between perfluoroalkyl substances (PFAS) and lipid concentrations in human populations. The results suggest that PFAS exposure is associated with higher levels of LDL, HDL, and TC, while the association with TG tends to be negative. Additionally, the study indicates a positive relationship between PFAS concentrations and cholesterol-related health outcomes. Further research is needed to investigate the effects of less-studied PFAS on human health.

State-of-the-art methods for exposure-health studies: Results from the exposome data challenge event

Summary: The exposome recognizes the simultaneous exposure of individuals to various environmental factors and aims to discover the factors behind diseases using a holistic approach. However, quantifying the health effects of complex exposure mixtures presents challenges. To address these challenges, the Barcelona Institute for Global Health organized a data challenge event where researchers from different disciplines could compete and apply state-of-the-art methods on a partially simulated exposome dataset.

Association between per and polyfluoroalkyl substances and markers of inflammation and oxidative stress

Ogbebor Enaholo Omoike et al.

Summary: The study revealed a significant association between Per and Polyfluoroalkyl substances (PFAS) and markers of chronic inflammation and oxidative stress. Increased exposure to PFAS led to higher serum concentration of these markers, indicating a connection between these chemicals and chronic inflammation and oxidative stress.

Per- and Polyfluoroalkyl Substance Toxicity and Human Health Review: Current State of Knowledge and Strategies for Informing Future Research

Suzanne E. Fenton et al.

Summary: Reports on the impacts of per- and polyfluoroalkyl substances (PFAS) on the environment and human health have increased substantially in peer-reviewed literature. Epidemiological studies have shown associations between exposure to specific PFAS and various health effects, and further research is needed to understand differences in responses between sexes, species, and life stages. More contemporary approaches such as read-across, molecular dynamics, and protein modeling are proposed to accelerate the development of toxicity information on PFAS, as well as the need for precaution to protect human health given the known effects of certain PFAS compounds.

Systemic PFOS and PFOA exposure and disturbed lipid homeostasis in humans: what do we know and what not?

Styliani S. Fragki et al.

Summary: The association between PFOS/PFOA and increased blood lipids in humans has been observed, while rodent studies show opposite effects, likely due to different exposure levels. Mechanistic research suggests that PFOS/PFOA activate the PPAR alpha pathway in human liver cells, but the involvement of other nuclear receptors is less clear. Future studies focusing on human-relevant test systems are needed to better understand the implications of PFOS/PFOA exposure on lipid homeostasis and human health.

Summary: This study used Gaussian graphical models to explore associations between environmental chemicals and endogenous molecules in a cohort of California women firefighters and office workers, revealing various exposure-metabolite associations and generating new hypotheses relevant to disease causation. Additionally, findings from this study were confirmed through analysis of data from the National Health and Nutrition Examination Survey, showing a novel approach to discovering links between chemical exposures and biological processes with potential implications for disease development.
