Role and architectural significance of porous chitosan-based scaffolds in bone tissue engineering

Effect of porosity on mechanical and biological properties of bioprinted scaffolds

Summary: The treatment of tissue defects is a major challenge due to donor shortage and issues with size, shape, and immunological rejection. This study investigates the effects of porosity percentage and pore shape on the mechanical and biological properties of porous polycaprolactone (PCL) scaffolds. The results demonstrate that scaffolds with a pore size of around 500 µm and porosity of 50% show the best cell proliferation after 14 days. These 3D bioprinted biodegradable scaffolds have potential applications in hard tissue engineering.

Chitosan-coated and thymol-loaded polymeric semi-interpenetrating hydrogels: An effective platform for bioactive molecule delivery and bone regeneration in vivo

Summary: In this study, a drug delivery vehicle was synthesized to enhance the bioactivity and sustained release of Thymol (Thy). The vehicle was made of semi-interpenetrating polymer network (SIPN) hydrogels containing sodium alginate and poly(2-ethyl-2-oxazoline), coated with chitosan. The results showed that this vehicle promoted osteoblast differentiation, activated the TGF-beta/BMP signaling pathway, and augmented bone regeneration.

Preparation of the bioglass/chitosan-alginate composite scaffolds with high bioactivity and mechanical properties as bone graft materials

Summary: Bioglass/chitosan-alginate (BCA) composite scaffolds with remarkable performance for bone tissue engineering were successfully prepared using freeze-drying method. The addition amount of sodium alginate (SA) was found to influence the microstructure, porosity, pore size, swelling ratio, degradation ratio, mechanical properties and mineralization ability of BCA composite scaffolds. The results showed that BCA composite scaffolds exhibited a three-dimensional interconnected network structure with high porosity and suitable pore size, and adding SA improved the mechanical strength and mineralization ability of BCA.

Novel non-toxic crosslinkers are being introduced to modulate release characteristics and achieve controlled drug release profiles for hydrogels made up of soluble and erodible polymers.
