Purification of monoclonal antibody using cation exchange z2 laterally-fed membrane chromatography - A potential alternative to protein A affinity chromatography

Summary: This study examines the purification of Trastuzumab using cation-exchange z(2) laterally-fed membrane chromatography (z(2)LFMC). The results show that the z(2)LFMC process is comparable to protein A affinity chromatography in terms of purity and recovery, but it is faster and does not require acidic conditions. The productivity of Trastuzumab with the z(2)LFMC process is more than 3 times higher than with column-based purification. Therefore, z(2)LFMC could be a cheaper and more efficient alternative to protein A affinity chromatography.

The kinetic mechanism of cations induced protein nanotubes self-assembly and their application as delivery system

Summary: In this study, the self-assembly process of carboxyl-abundant enzymolyzed alpha-lactalbumin was investigated using a step-wise method. It was found that Ca2+ played a crucial role in the formation of nanotubes. The study also demonstrated the potential of alpha-lactalbumin nanotubes as a delivery system for food applications, as they successfully encapsulated curcumin and improved the viscosity of liquid food.

The results showed that treatment with melatonin for 20 days did not affect ovarian functions or the response to an estrous synchronization treatment regimen and TAI.

Engineering of the unfolded protein response pathway in Pichia pastoris: enhancing production of secreted recombinant proteins

Summary: Protein folding and processing in the endoplasmic reticulum can hinder protein production and secretion in Pichia pastoris. The unfolded protein response pathway is activated in response to misfolded proteins, upregulating chaperones and folding-assisting proteins. Co-expression of specific UPR genes with the recombinant gene of interest is a common strategy to enhance protein production, but can lead to decreased productivity if not balanced properly.

Efficient Secretory Expression and Purification of Food-Grade Porcine Myoglobin in Komagataella phaffii

Summary: In this study, secretory expression of porcine myoglobin was achieved by engineered Komagataella phaffii, with optimized fermentation conditions resulting in the highest titer of 285.42 mg/L in fed-batch fermentations. Additionally, a purification method for food-grade myoglobin was developed, with a purity of 88.0% and a recovery rate of 66.1% obtaining 0.22 mol of heme/mol of myoglobin.

Recombination machinery engineering facilitates metabolic engineering of the industrial yeast Pichia pastoris

Summary: A CRISPR-Cas9 based genome editing platform was established, significantly enhancing the recombination efficiency of P. pastoris and allowing for efficient seamless gene disruption and multiple gene assembly. Additionally, this tool resulted in a 30-fold increase in fatty alcohol production and enabled regulation of gene expression and metabolic pathways under different growth conditions.
