Curcumin in human osteosarcoma: From analogs to carriers

Summary: Osteosarcoma, the most prevalent bone tumor in adolescents, has low survival rates in patients with metastatic disease. To improve the clinical applications of curcumin, potential curcumin analogs and nanobased curcumin delivery systems have been developed. This critical review focuses on the characteristics of curcumin and its analogs, as well as strategies to improve their bioactivity and bioavailability, especially in the treatment of metastatic osteosarcoma. The review also highlights promising nanoformulations and scaffold systems for the targeted delivery of curcumin in human osteosarcoma cells.

Summary: Nanoemulsion encapsulated in ionotropic hybrid hydrogel beads of alginate, chitosan, gelatin and polyethylene oxide can effectively deliver hydrophobic active ingredients like curcumin, enhancing their bioavailability and biological activities. The release behavior in simulated gastric and intestinal fluids showed faster release in gastric fluid, potentially due to mesh size, hydration extent, and interaction with Ca2+ ions. The released curcumin in gastric fluid showed significantly greater free radical scavenging and antibacterial activities compared to intestinal fluid.

Curcumin-loaded composite hydrogel based on scallop (Patinopecten yessoensis) male gonad hydrolysates and κ-carrageenan: Characterization and in vitro digestibility

Summary: A composite hydrogel consisting of scallop male gonad hydrolysates (SMGHs) and kappa-carrageenan (KC) was successfully fabricated with a designed microstructure to protect curcumin (Cur) and exhibited a more compact network structure, leading to a slower release rate and a final release extent exceeding 90%. The hydrogels, adaptable to the intestinal environment, were identified as reliable and desirable soft materials for embedding and delivering Cur to the colon, showing potential applications in functional foods and biological therapeutic purposes.

Summary: In this study, pH-responsive GG-MAA-AMPS hydrogel delivery system was successfully prepared and characterized. The hydrogels exhibited good drug loading and release behavior, and showed safety in oral tolerability test. These findings suggest that GG-MAA-AMPS hydrogel could be a potential carrier for metformin HCl and other hydrophilic drugs.
