Characterization of degradation patterns and enzymatic properties of a novel alkali-resistant alginate lyase AlyRm1 from Rubrivirga marina

Summary: This study successfully cloned and expressed the PL6 alginate lyase gene alyrm1 from Rubrivirga marina. The recombinant AlyRm1 showed good activity and stability at 30 degrees C and pH 10.0, and it could preferentially degrade into AOSs with DP 2-5 and monosaccharides, suggesting its potential applications.

Human gut bacteria can utilize macroalgal glycans to produce oligosaccharides, which interact with immune cell receptors and are available for microbial fermentation, playing important roles in balancing gut homeostasis. However, further research is needed to understand the molecular interactions between glycans and bacteria/cells.

A Novel Alginate Lyase: Identification, Characterization, and Potential Application in Alginate Trisaccharide Preparation

Summary: Alginate oligosaccharides (AOS) have various biological activities and important applications, and alginate lyases have unique advantages in the preparation of AOS. In this study, a strain of Pseudoalteromonas arctica M9 was isolated from Sargassum, and five putative alginate lyases were identified in its genome. Among them, AlyM2 showed the highest activity and mainly produced trisaccharides from sodium alginate, indicating its potential for trisaccharide preparation.

Improving the thermostability of alginate lyase FlAlyA with high expression by computer-aided rational design for industrial preparation of alginate oligosaccharides

Summary: This study reports the enhancement of thermostability of FlAlyA, an alginate lyase with industrial potential. By computer-aided calculation and site-directed mutagenesis, two single-point mutants with better thermostability were optimized and the mechanisms of these mutants were investigated. The expression level of the mutants was greatly increased, making them potential candidates for the industrial production of alginate oligosaccharides.
