SARS-CoV-2 spike protein as a bacterial lipopolysaccharide delivery system in an overzealous inflammatory cascade

Summary: Accumulating evidence suggests that bacterial lipopolysaccharide (LPS) plays a potential role in the excessive immune response during SARS-CoV-2 infection. LPS binds to the SARS-CoV-2 spike (S) protein, enhancing proinflammatory effects. LPS interacts with multiple hydrophobic pockets on both the S1 and S2 subunits of the S protein. The binding affinity of LPS to the S2 pocket is lower than that to the S1 pocket, indicating its role as an intermediate in LPS transfer. This molecular mechanism explains how the S protein enhances LPS-mediated hyperinflammation.
