Activation of TLR4 induces severe acute pancreatitis-associated spleen injury via ROS-disrupted mitophagy pathway

E. Wen et al.

Summary: This study investigated the mechanism of spleen injury caused by severe acute pancreatitis (SAP) and explored the potential use of tuftsin in treating spleen damage. The researchers found that the up-regulation of Toll-like receptor 4 (TLR4) in the spleen leads to the accumulation of reactive oxygen species (ROS) and mitochondrial dysfunction under SAP conditions. They also discovered that N-acetylcysteine (NAC) treatment or knocking out TLR4 can improve splenic ROS and mitochondrial dysfunction in SAP mice. Furthermore, they verified that spleen injury is caused by splenic ROS affecting the PI3K/p-AKT/mTOR pathway-mediated autophagy. The study also showed that tuftsin supplementation can ameliorate the pathological damage, ROS accumulation, impaired autophagy, inflammation expression, and apoptosis in damaged spleen.

Gene Expression Profiling: Identification of Novel Pathways and Potential Biomarkers in Severe Acute Pancreatitis

Maryam Nesvaderani et al.

Summary: This study identified genes and molecular mechanisms associated with the pathogenesis of severe acute pancreatitis (AP) by analyzing global gene expression profiles. Specific immunological pathways were found to play a central role in the severity of AP. A four-gene transcriptomic signature was also determined that can predict severe AP with an accuracy of 64%.

Toll-Like Receptor Signaling and Its Role in Cell-Mediated Immunity

Tianhao Duan et al.

Summary: This article provides a detailed overview of the innate immune system's defense mechanisms against pathogens, with a focus on the role of Toll-like receptors (TLRs) in immune responses. TLRs not only link innate immunity to adaptive immunity, but also directly regulate the function of T cells. In addition, the article discusses the critical role of TLR signaling in host defense against infectious diseases, autoimmune diseases, and cancer.

Identification of novel immune-related targets mediating disease progression in acute pancreatitis

Qiang Liu et al.

Summary: This study found that SIRS and CARS are simultaneously activated during the progression of AP. Additionally, it discovered that TLR2 may be a novel therapeutic target for regulating dysregulation of inflammatory responses during AP progression.

Kinsenoside alleviates inflammation and fibrosis in experimental NASH mice by suppressing the NF-ΚB/NLRP3 signaling pathway

Yan-fang Deng et al.

Summary: This study reveals that kinsenoside (KD) can alleviate non-alcoholic steatohepatitis (NASH) by inhibiting the NF-Kappa B/NLRP3 signaling pathway. KD shows hepatoprotective properties and has the potential to be a novel and effective drug for treating NASH.

Effect of emodin on long non-coding RNA-mRNA networks in rats with severe acute pancreatitis-induced acute lung injury

Caiming Xu et al.

Summary: This study investigated the therapeutic effects of emodin and DEX on SAP-ALI, revealing that both drugs were effective. Differences in mRNA and lncRNA levels were observed between emodin- and DEX-treated SAP-ALI rats, with functional analysis showing enrichment in inflammatory and immune response pathways for emodin. Emodin may be considered as a suitable alternative or complementary medicine for SAP-ALI treatment.

The Role of Pancreatic Infiltrating Innate Immune Cells in Acute Pancreatitis

Cheng Peng et al.

Summary: Acute pancreatitis is a leading cause of gastrointestinal-related hospital admissions with significant morbidity and mortality. Immune cells play a critical role in the pathogenesis of AP, and immunomodulatory therapy targeting them may improve patient prognosis. Cells of the innate immune system, including macrophages and neutrophils, are the majority of infiltrating cells during AP.
