Microglial depletion exacerbates motor impairment and dopaminergic neuron loss in a 6-OHDA model of Parkinson's disease

Carolina Parga Martins Pereira et al.

Summary: 6-hydroxydopamine is a neurotoxin commonly used to induce Parkinson's disease in mice. It has been found that microglia may have contradictory effects in PD, either exerting neuroprotection or exacerbation of neuronal death. This study highlights the protective role of microglia in dopaminergic neuron survival during the initial phase of PD, and further investigation of the mechanisms involved will contribute to a better understanding of the pathophysiology of PD.

Role of the AMPA receptor in antidepressant effects of ketamine and potential of AMPA receptor potentiators as a novel antidepressant

Atsushi Suzuki et al.

Summary: Ketamine has rapid and long-lasting antidepressant effects, but its clinical use is limited by its undesirable side effects. This review focuses on the discovery of a novel AMPA-R potentiator, TAK-653, with low agonistic activity, which may overcome the limitations of known AMPA-R potentiators. The study highlights the importance of AMPA-R activation in mediating the antidepressant effects of ketamine.

The complex clinical response to selective serotonin reuptake inhibitors in depression: a network perspective

Lynn Boschloo et al.

Summary: This study aimed to determine the direct and indirect effects of SSRIs on clinical symptoms of depression. The results showed that SSRIs primarily reduced affective symptoms, which were related to reductions in cognitive symptoms and some specific arousal/somatic symptoms. These findings contribute to understanding the mechanisms of SSRIs and can facilitate early detection of treatment responders and non-responders in clinical practice.

ATP-mediated signalling in the central synapses

Ulyana Lalo et al.

Summary: ATP released from synaptic terminals and astrocytes can activate neuronal P2 receptors, contributing to slow and diffuse modulation of synaptic dynamics and neuronal firing in the CNS. Neuronal P2X and P2Y receptors can be activated by ATP released from synaptic terminals, astrocytes, and microglia, participating in the regulation of synaptic homeostasis and plasticity. Purinergic regulation of synaptic transmission plays an important role in various physiological and pathological contexts.

Implications for the role of lipopolysaccharide in the development of atherosclerosis

Armita Mahdavi Gorabi et al.

Summary: Mounting scientific evidence has established that atherosclerosis is a chronic inflammatory disorder, and one potential source of vascular inflammation is the presence of pathogenic bacteria, particularly lipopolysaccharide (LPS). Mutations in Toll-like receptor (TLR)-4, which is involved in recognizing bacterial LPS, have been found in various ethnic groups and are associated with a decreased risk of atherosclerosis. Epidemiological studies have also suggested that LPS is a risk factor for the development of atherosclerosis.

Microglia in depression: an overview of microglia in the pathogenesis and treatment of depression

Haixia Wang et al.

Summary: This review highlights the role of microglia in the pathology of major depressive disorder. It discusses microglial activation in animal models and clinically depressed patients, as well as the mechanisms by which microglia recognize depression-associated stress and regulate conditions. The review also describes the effects of antidepressants on microglial activation.

Summary: MDD is a common psychiatric disorder with significant impact on individuals and the economy. Chronic stress is a major environmental risk factor for depression, with women being more susceptible. Rodent models of depression focusing on susceptibility and resilience provide valuable insights into the mechanisms underlying the disease.

Intranasal lipopolysaccharide administration prevents chronic stress-induced depression- and anxiety-like behaviors in mice

Qun Lu et al.

Summary: Intranasal administration of LPS prior to stress exposure can prevent chronic social defeat stress-induced depression and anxiety-like behaviors, as well as reduce neuroinflammatory responses in the hippocampus and prefrontal cortex. This suggests that innate immune stimulation via intranasal administration could be a potential approach for the prevention of depression and anxiety.

Specific depletion of resident microglia in the early stage of stroke reduces cerebral ischemic damage

Ting Li et al.

Summary: The specific depletion of microglia in the early stage of ischemic stroke can significantly reduce the infarct volume and improve motor ability. Depleting microglia also leads to a decrease in degenerating neurons and inducible nitric oxide synthase positive (iNOS(+)) cells. Moreover, the depletion of microglia results in an increase in anti-inflammatory factors and a decrease in pro-inflammatory factors 3 days after stroke. These findings suggest that modulation of the inflammatory response through the elimination of microglia may be a promising therapeutic approach for the treatment of cerebral ischemia.
