Radiation-induced senescence: therapeutic opportunities

Summary: The limitation of cancer radiotherapy lies in the challenge of destroying tumors without causing excessive damage to critical tissues and organs and compromising patient's quality of life. Cellular senescence induced by radiation in both tumor and adjacent normal tissues contributes to tumor recurrence, metastasis, and therapy resistance, while chronic senescent cells in normal tissues and organs lead to late damaging effects. This review discusses the identification of cellular senescence through biomarkers and the role of senescence-associated secretory phenotype in radiation-induced late effects. Therapeutic options to eliminate cellular senescence using senolytics and/or senostatics are also explored, along with the potential of cellular reprogramming to enhance the effectiveness of radiotherapy.

Anisodamine potently inhibits SARS-CoV-2 infection in vitro and targets its main protease

Summary: This study found that anisodamine suppressed SARS-CoV-2 infection in Vero E6 cells and reduced SARS-CoV-2 pseudovirus entry in HEK293/hACE2 cells. Molecular docking studies suggested that anisodamine may target SARS-CoV-2 main protease (Mpro) with a docking score of -6.63 kcal/mol and form three hydrogen bonds at the predicted active site of Mpro.

Experimental results showed that this radio-protective agent could alleviate radiation-induced toxicities through mechanisms such as anti-oxidant, antiapoptosis, and anti-inflammatory effects. Co-treatment of alpha-lipoic acid with radiotherapy could ameliorate the radiation-induced toxicities in healthy cells/tissues.
