Anti-inflammation and anti-aging mechanisms of mercaptopurine in vivo and in vitro

HuiJie Jia et al.

Summary: This study found that mercaptopurine inhibits cellular and animal senescence, and exhibits anti-inflammatory effects in UV-B-induced skin damage and photoaging. It achieves this by suppressing important signaling molecules MAPK and NF-kB, increasing collagen fiber content, and facilitating collagen synthesis, thereby protecting the skin from UV-B damage.

Pancreatic Cancer and Cellular Senescence: Tumor Microenvironment under the Spotlight

Michela Cortesi et al.

Summary: Pancreatic ductal adenocarcinoma (PDAC) has a poor prognosis due to immune suppression and the tumor microenvironment. This review explores the interplay between senescent and non-senescent cell types and their impact on PDAC progression. The non-tumoral cells in the tumor microenvironment influence key aspects of tumor growth, metabolism, cell death, and treatment resistance. Understanding these interactions is crucial for PDAC treatment.

Effect of acute high-intensity exercise on myocardium metabolic profiles in rat and human study via metabolomics approach

Lijun Wu et al.

Summary: This study aimed to explore the effects of acute high-intensity exercise on rat myocardial metabolism and evaluate its diagnostic potential for sports injuries. It was found that acute high-intensity exercise caused changes in myocardial metabolism, including disorders in complex lipids and fatty acids, an increase in purine degradation products, and signs of impaired glucose metabolism. Additionally, amino acids were enhanced and exerted a protective effect on the myocardium.

Tea Polyphenols as Prospective Natural Attenuators of Brain Aging

Mengyu Hong et al.

Summary: Aging is inevitable and often accompanied by chronic disease. Tea polyphenols have potential anti-aging benefits by regulating the composition of the intestinal microbiota. The interaction between human neurological function and the gut microbiota suggests that tea polyphenols can be an effective intervention to alleviate brain-aging.

Study on Huangqi Bazhen Decoction on Relieving Chemotherapy Intestinal Mucositis in Capecitabine Gavage Mice

Tieyi Shi et al.

Summary: This study investigated the therapeutic effect and mechanism of Huangqi Bazhen decoction on chemotherapeutic intestinal mucositis induced by capecitabine in mice, in order to provide evidence for clinical application. The experimental results showed that Huangqi Bazhen decoction reversed the morphological features of intestinal injury and significantly reduced weight loss and diarrhea during capecitabine treatment. Furthermore, it was found that Huangqi Bazhen decoction improved antioxidant enzyme levels and significantly reduced inflammation markers.

Network analysis and experimental pharmacology study explore the protective effects of Isoliquiritigenin on 5-fluorouracil-Induced intestinal mucositis

Yi-fan Liao et al.

Summary: This study investigated the impact of isoliquiritigenin (ISL) on 5-fluorouracil (5-FU)-induced intestinal mucositis. Network analysis and molecular docking were used to predict the targets and mechanism of ISL, which were then validated in a mouse model. The results suggested that ISL could attenuate 5-FU-induced intestinal mucositis by regulating the TNF/NF-κB pathway and inhibiting inflammatory mediators PTGS2 and NOS2.

Exploiting senescence for the treatment of cancer

Liqin Wang et al.

Summary: This review discusses how senescence can be induced in cancer cells and how the distinctive features of senescent cancer cells can be exploited for selective eradication. It also explores activation of the host immune system as an attractive way to clear senescent cancer cells, and the potential benefits of a sequential treatment approach with pro-senescence therapy followed by senolytic therapy.

Senolytic Combination of Dasatinib and Quercetin Alleviates Intestinal Senescence and Inflammation and Modulates the Gut Microbiome in Aged Mice

Tatiana Dandolini Saccon et al.

Summary: The study found that the senolytic drug combination Dasatinib plus Quercetin (D+Q) significantly reduces senescent cell and inflammatory burden in the intestines of aged mice, while also altering specific microbiota signatures. The most prominent changes were observed in the small intestine. This suggests that optimized senolytic regimens may improve health in older subjects by reducing intestinal senescence, inflammation, and microbial dysbiosis.

The Era of Janus Kinase Inhibitors for Inflammatory Bowel Disease Treatment

Jin-Woo Kim et al.

Summary: Treatment response in inflammatory bowel disease (IBD) remains a significant issue, with anti-tumor necrosis factor therapies and Janus kinase inhibitors offering potential solutions. While effective in ulcerative colitis (UC), Janus kinase inhibitors show less efficacy in Crohn's disease (CD). Safety concerns persist due to the immune-system-based mechanism of these drugs.

Therapeutic Potential for Regulation of the Nuclear Factor Kappa-B Transcription Factor p65 to Prevent Cellular Senescence and Activation of Pro-Inflammatory in Mesenchymal Stem Cells

Rocio Mato-Basalo et al.

Summary: Mesenchymal stem cells have potential in treating age-related diseases, and small extracellular vesicles derived from these cells have been proposed as therapies. The activation of p65 is found to be important in cellular senescence and inflammatory activation in mesenchymal stem cells. Inhibiting p65 can prevent cellular senescence and the activation of proinflammatory molecules, highlighting its role in restoring functionality and preventing inflamm-aging.

Therapy-Induced Senescence: Opportunities to Improve Anticancer Therapy

Pataje G. Prasanna et al.

Summary: Cellular senescence is crucial in suppressing tumor growth, but radiation and chemotherapy can lead to accumulation of senescent cells in both tumor and normal tissues, potentially promoting tumor relapse and resistance to treatment. David Dahlgren et al.

Summary: Chemotherapeutics-induced intestinal mucositis (CIM) is a common gastrointestinal toxicity in cancer patients due to the rapid proliferation of intestinal epithelial cells and complex immunological interactions with gut microbiota. CIM often leads to dose-reduction, decreasing the efficacy of anticancer treatment, and effective strategies for prevention and treatment are still lacking.
