Respiratory distress in small animals: Pathophysiology and clinical approach

Jennifer M. Loewen et al.

Summary: Understanding respiratory physiology is crucial for accurately identifying the source of respiratory distress and implementing appropriate therapies. Prompt recognition of unique clinical signs is important to localize the disease process and provide targeted treatment. Different diseases affecting distinct parts of the respiratory tract lead to distinguishing clinical signs.

BAY11-7082 inhibits the expression of tissue factor and plasminogen activator inhibitor-1 in type-II alveolar epithelial cells following TNF-α stimulation via the NF-κB pathway

Yumei Cheng et al.

Summary: The study demonstrates that the NF-κB pathway inhibitor BAY11-7082 improves hypercoagulation and fibrinolytic inhibition in alveolar epithelial cells induced by TNF-alpha by suppressing the expression of TF and PAI-1.

Runx1 is a central regulator of osteogenesis for bone homeostasis by orchestrating BMP and WNT signaling pathways

Chen-Yi Tang et al.

Summary: Runx1 plays a crucial role in osteogenesis by regulating BMP and Wnt signaling pathways. Knockout of Runx1 disrupts bone homeostasis by decreasing osteogenesis and increasing adipogenesis. The findings highlight the potential of targeting Runx1 for novel therapeutics in osteoporosis.

Prone Positioning in Moderate to Severe Acute Respiratory Distress Syndrome Due to COVID-19: A Cohort Study and Analysis of Physiology

Mehdi C. Shelhamer et al.

Summary: Prone positioning in patients with moderate to severe ARDS due to COVID-19 is associated with reduced mortality and improved physiologic parameters, with one in-hospital death averted for every 8 patients treated. The results suggest that prone positioning may represent an additional therapeutic option in patients with ARDS due to COVID-19, but further replication and scaling of the intervention are important.

Pulmonary Procoagulant and Innate Immune Responses in Critically Ill COVID-19 Patients

Esther J. Nossent et al.

Summary: This study evaluated the activation of host response pathways in the lung compartment of critically ill COVID-19 patients with persistent acute respiratory distress syndrome. The results showed strong local responses relating to coagulation, the complement system, cytokines, chemokines and growth factors in the bronchoalveolar compartment, with some biomarkers declining in levels at 3 to 4 weeks after mechanical ventilation initiation.

Andrographolide sulfonate attenuates alveolar hypercoagulation and fibrinolytic inhibition partly via NF-xB pathway in LPS-induced acute respiratory distress syndrome in mice

Hong Qian et al.

Summary: Andro-S dose-dependently inhibited lung tissue factor (TF) and plasminogen activator inhibitor (PAI)-1 expressions in LPS-induced ARDS mice, while also decreasing TF, PAI-1, thrombin-antithrombin complex (TAT), procollagen peptide type III (PIIIP) concentrations and promoting activated protein C (APC) production in bronchoalveolar lavage fluid (BALF). Andro-S effectively inhibited LPS-induced inflammatory response and inactivated NF-xB pathway.

Personalized mechanical ventilation in acute respiratory distress syndrome

Paolo Pelosi et al.

Summary: A personalized mechanical ventilation approach based on lung physiology, ARDS etiology, lung imaging, and biological phenotypes may improve ventilation practice and outcomes for ARDS patients, but further research is needed before it can be applied clinically. Ventilatory parameters should be titrated based on physiological variables and individualized goals, with a focus on low tidal volume and driving pressures.
