Summary: In this study, the role of DKK3 and its interplay with microglial activation in neuropathic pain were investigated. The results suggest that DKK3 ameliorates neuropathic pain by inhibiting ASK-1/JNK/p-38-mediated microglia polarization and neuroinflammation, at least in part, through the Kremen-1 and DVL-1 pathways.

Summary: In this study, the preventive effects of intrathecal bupivacaine administration against neuropathic pain were investigated. Bupivacaine attenuated pain behaviors and inflammatory alternations by suppressing microglia M1 polarization and pro-inflammatory cytokine expression. The Interferon Regulatory Factor 5 (IRF5)/P2X Purinoceptor 4 (P2X4R) and High Mobility Group Box 1 (HMGB1)/Toll-Like Receptor 4 (TLR4)/NF-kappa B inflammatory axes were identified as key players in microglia polarization and cytokine expression.

Expression Profiling of L5-S2 Spinal Cord Dorsal Horn in a Rat Model of Chronic Pelvic Pain Syndrome Uncovers Potential Mechanism of Electroacupuncture Mediated Inflammation and Pain Responses

Summary: The purpose of this study was to explore the expression profiles of genes in the spinal cord dorsal horn (SCDH) of a chronic pelvic pain syndrome (CPPS) rat model that are relevant to pain and inflammation using RNA-Seq, and to investigate the mechanism of the anti-inflammatory and analgesic effects of electroacupuncture (EA). The results showed that CPPS rats exhibited persistent mechanical allodynia and severe tissue inflammatory reactions. After EA therapy, the pain sensitivity and inflammatory response of CPPS rats decreased significantly.

Summary: PD-L1 is significantly upregulated after SCI and highly expressed on macrophages/microglia at the injury epicenter. Deletion of PD-L1 results in worse locomotor recovery and more serious pathological pain, which is associated with increased polarization of M1-like macrophages/microglia. Mechanistic analysis suggests that PD-L1 may improve functional outcomes following SCI by inhibiting phosphorylation of p38 and ERK1/2.
