Effects of Ruxolitinib on fibrosis in preclinical models of systemic sclerosis

Summary: Systemic sclerosis (SSc) is an autoimmune fibrotic disorder characterized by antinuclear autoantibody production and excessive apoptotic cells. This study investigated the potential of a JAK inhibitor, ruxolitinib, to improve efferocytosis and fibrosis in SSc. Results showed that ruxolitinib reduced dermal thickness, fibrosis, and caspase-3 positive cells in a SSc mouse model. Ruxolitinib also improved lung fibrosis and efferocytosis capacity of macrophages in vitro. Furthermore, ruxolitinib decreased TGF-beta activation and phosphorylation of SMAD2/3 and STAT3 in human lung fibroblasts. Overall, this study supports the anti-fibrotic properties of ruxolitinib in SSc.

Synergistic Effects of Quercetin-Modified Silicone Gel Sheet in Scar Treatment

Summary: Both silicone gel and quercetin are effective in scar treatment, but their mechanisms of action differ. This study successfully developed a silicone gel sheet that releases quercetin in a sustained manner. The combination of quercetin-modified silicone gel sheet was found to be more effective in inhibiting cell proliferation in scar tissue than either treatment alone.

The role of chest CT in deciphering interstitial lung involvement: systemic sclerosis versus COVID-19

Summary: This study aimed to differentiate between COVID-19 pneumonia and SSc-ILD using CT features. Key findings included fibrosis inside focal ground-glass opacities in the upper lobes for SSc-ILD and consolidation in the lower lobes for COVID-19 pneumonia. A predictive score was developed with high sensitivity and specificity for COVID-19 diagnosis.

SSC-ILD mouse model induced by osmotic minipump delivered bleomycin: effect of Nintedanib

A model using bleomycin (BLM) infusion in mice was used for drug screening, with Nintedanib (NINT) showing anti-fibrotic effects on lung but not skin.
