Inhibition of interleukin-6 trans-signaling improves survival and prevents cognitive impairment in a mouse model of sepsis

Summary: Sepsis-associated encephalopathy (SAE) is characterized by acute and chronic cognitive impairments and is associated with increased morbidity and mortality. This study investigated the potential therapeutic target of inhibiting IL-6 trans-signalling for sepsis and SAE. The results showed that selective inhibition of IL-6 trans-signalling by sgp130 exerted protective effects against SAE in a mouse model of sepsis.

Increased interleukin-6 levels associated with malaria infection and disease severity: a systematic review and meta-analysis

Summary: This systematic review and meta-analysis compared IL-6 levels between different groups of malaria patients and healthy controls. The results showed significantly higher levels of IL-6 in patients with severe malaria compared to those with non-severe malaria, as well as in patients with uncomplicated malaria compared to the control group. However, no significant differences in IL-6 levels were found between patients with uncomplicated malaria and asymptomatic malaria, or between patients with asymptomatic malaria and healthy controls. Higher IL-6 levels were observed in patients who died compared to those who survived. These findings are important for monitoring and studying the severity of malaria.

Summary: This study investigates the role and regulation of MDSCs in the context of malaria. The results show that MDSCs expand in the spleen and bone marrow during Experimental Cerebral Malaria (ECM) and exhibit suppressive properties and high production of Nitric oxide (NO). Depletion of MDSCs regulates immune balance and inflammatory changes. IL-6 plays an important role in the expansion of MDSCs and expression of inflammatory markers.

Summary: Pulmonary emphysema is associated with dysregulated innate immune responses. In this study, the authors investigated the role of innate immune inflammasome complexes in promoting emphysema. They found that ASC, Caspase-1, and AIM2 were up-regulated in emphysematous patients and mouse models of emphysema. AIM2 was associated with elevated levels of IL-1 beta. Blockade of AIM2 and IL-1 receptor ameliorated emphysema in mice. IL-6 trans-signaling was also found to augment AIM2 expression in emphysema.
