Endothelial PDGF-BB/PDGFR-β signaling promotes osteoarthritis by enhancing angiogenesis-dependent abnormal subchondral bone formation

Summary: The mechanisms that coordinate the shift from joint homeostasis to osteoarthritis (OA) remain unknown. Accumulating evidence suggests that subchondral bone deterioration may trigger cartilage degeneration. This study found that the expression of PDGFR-beta, mainly in CD31(hi)Emcn(hi) endothelium, was elevated in subchondral bones from OA patients and rodent OA models. A mouse model with PDGFR-beta deletion in endothelial cells showed reduced H-type vessels and improved subchondral bone deterioration and cartilage degeneration. Inhibiting PDGFR-beta in subchondral bone through local injection effectively attenuated the pathogenesis of OA. Targeting PDGFR-beta could be a novel approach for preventing or treating OA.
