Blocking Cx43 alleviates neuropathic pain in rats with chronic constriction injury via the P2X4 and P38/ERK-P65 pathways

Juping Xing et al.

Summary: This study investigated the role of Cx43 in neuropathic pain mediated by the P2X4 receptor in rats with chronic constriction injury (CCI). The results showed that blocking Cx43 can alleviate neuropathic pain in CCI rats through the P2X4, p38, ERK, and NF-kB signaling pathways. These findings suggest that Cx43 may be a promising therapeutic target for the development of novel pharmacological agents in the management of neuropathic pain.

The Purinergic Receptor P2X4 Promotes Th17 Activation and the Development of Arthritis

Chakib Hamoudi et al.

Summary: This study found that the purinergic receptor P2X4 is associated with human Th17 cells and is required for their development and IL-17 production. Inhibition of P2X4 receptor does not affect Th1 cells and IFN-gamma production. This study provides insights into the role of purinergic signaling in T cell activation and suggests P2X4 as a critical factor in Th17 activation and autoimmune arthritis.

Eleonore Bertin et al.

Summary: Increased surface density of neuronal P2X4 contributes to synaptic deficits, alterations in anxiety and memory functions, consistent with the implication of P2X4 in neuropsychiatric and neurodegenerative disorders. These conditional P2X4mCherryIN knock-in mice provide a new approach to explore the cell-specific roles of P2X4 in various physiological and pathological contexts.

Fast green FCF inhibits Aβ fibrillogenesis, disintegrates mature fibrils, reduces the cytotoxicity, and attenuates Aβ-induced cognitive impairment in mice

Fufeng Liu et al.

Summary: This study found that Fast Green FCF (FGF) can effectively inhibit Aβ-induced neurotoxicity by reducing Aβ fibrillogenesis, disintegrating mature fibrils, decreasing Aβ-induced cytotoxicity, and improving Aβ-induced learning and memory impairments. Molecular dynamics simulations demonstrated that FGF can interact with Aβ through various mechanisms to reduce its neurotoxic effects.

The food additive fast green FCF inhibits α-synuclein aggregation, disassembles mature fibrils and protects against amyloid-induced neurotoxicity

Fenghua Wang et al.

Summary: The food additive fast green FCF was found to inhibit alpha-synuclein fibrillogenesis and reduce cytotoxicity by disrupting the pentamer structure, indicating its potential as a drug candidate against amyloid fibril aggregation in Parkinson's disease. The interaction mechanism between fast green FCF and alpha-synuclein was elucidated through molecular dynamics simulations, revealing the disruption of pentamer structure and reduction of beta-sheet content. Electrostatic interactions, hydrogen bonds, and pi-pi interactions were identified as key factors in the binding of fast green FCF to alpha-synuclein.
