Selective, Disruptive Luminescent Ru(II) Polypyridyl Probes of G-Quadruplex

Summary: This study reports a Ru(II) complex (Ru-PDC3) that combines the light switch capability of a Ru(II) dipyridylphenazine complex with the well-known G4-selective ligand Phen-DC3 into a single structure. The complex exhibits the expected light switch effect and strong affinity for G4 structures, while also possessing a powerful G4 disrupting capability. Additionally, the complex is fully water-soluble and readily permeable to live cells.

Copper-Nitrogen-Coordinated Carbon Dots: Transformable Phototheranostics from Precise PTT/PDT to Post-Treatment Imaging-Guided PDT for Residual Tumor Cells

Summary: Phototheranostics is a promising approach for cancer diagnosis and treatment, but the limitations of laser irradiation make it challenging to completely eradicate solid tumors. This study developed a novel transformable phototheranostics using copper-nitrogen-coordinated carbon dots assembled with hyaluronic acid. The transformable nanoplatform specifically targets CD44-overexpressing tumor cells and enables precise photothermal therapy and photodynamic therapy. The transformation of the nanoplatform in tumor cells allows for post-treatment imaging-guided therapy on residual tumor cells.

Near-infrared-II-activatable sulfur-deficient plasmonic Bi2S3-x-Au heterostructures for photoacoustic imaging-guided ultrasound enhanced high performance phototherapy

Summary: In this study, an all-in-one phototheranostic nanoplatform, Bi2S3-x-Au@HA, was successfully synthesized for photoacoustic (PA) imaging-guided sonodynamic therapy (SDT)-enhanced photodynamic therapy (PDT)/photothermal therapy (PTT). Through density functional theory analysis, it was confirmed that light and ultrasound-induced electrons are more likely to aggregate on the Au nanoparticle surface through interfacial self-assembly, promoting electron-hole separation and enhancing photocatalytic activity with increased reactive oxygen species (ROS) generation. With the modification of hyaluronic acid (HA), Bi2S3-x-Au@HA can selectively target cancer cells through HA and CD44 protein interactions. In both in vitro and in vivo experiments, Bi2S3-x-Au@HA effectively suppressed tumor growth under a single NIR-II laser and ultrasound irradiation with negligible toxicity. Our findings provide a framework for fabricating Schottky-type heterostructures as single NIR-II light-responsive nanotheranostic agents for PA imaging-guided cancer phototherapy.

Ru(II) containing photosensitizers for photodynamic therapy: A critique on reporting and an attempt to compare efficacy

Summary: Ruthenium(II)-based coordination complexes have great potential as photosensitizers for photodynamic therapy. The modular architectures of these complexes allow for tuning of various properties such as cellular uptake, subcellular targeting, solubility, and light absorption. Many studies focus on systematic variation of ligands to investigate their impact on photophysical and photobiological performance. However, comparisons between Ru(II)-based PSs and other generations of PSs are scarce. Nonetheless, quantitative models based on PDT dose metrics are available for qualitative comparisons. This review summarizes the relevant studies on Ru(II)-containing PSs and their potential efficacy.

Black SnO2-x based nanotheranostic for imaging-guided photodynamic/photothermal synergistic therapy in the second near-infrared window

Summary: This study developed nanotheranostic agents based on oxygen-deficient black SnO2-x and hyaluronic acid (HA) for combined tumor treatment in the NIR-II window. The nanotheranostic agents showed high dispersity and biocompatibility, allowing specific targeting of tumor sites and simultaneous generation of sufficient hyperthermia and reactive oxygen species. The synergistic photodynamic therapy (PDT) and photothermal therapy (PTT) with targeted specificity significantly inhibited tumor growth under NIR-II light irradiation guidance.

Photoinduced DNA Cleavage and Photocytotoxic of Phenanthroline-Based Ligand Ruthenium Compounds
