Radiochemical Quality Control Methods for Radium-223 and Thorium-227 Radiotherapies

Stable Chelation of the Uranyl Ion by Acyclic Hexadentate Ligands: Potential Applications for 230U Targeted α-Therapy

Summary: This study investigated four equatorial-spanning ligands as uranium chelators. The results showed that the complexes formed by these ligands with UO22+ were highly stable and exhibited good stability in vivo. The hox ligand demonstrated higher thermodynamic stability, likely due to its lower strain energy. The ligands remained >95% stable in human plasma and >80% intact in the presence of hydroxyapatite (HAP). In vivo experiments in mice showed that the uranium complexes of these ligands did not accumulate in the skeletal system. Overall, these equatorial-spanning ligands hold great promise for 230U TAT.

Glypican-3-Targeted 227Th α-Therapy Reduces Tumor Burden in an Orthotopic Xenograft Murine Model of Hepatocellular Carcinoma

Luminescence studies indicate the presence of water molecules in the inner metal coordination spheres.

Summary: Both [Zr-89]Zr-3,2-HOPO-MSLN-mAb and [Zr-89]Zr-DFO-MSLN-mAb showed low nanomolar binding affinity for MSLN and >95% immunoreactivity. In vivo studies demonstrated that [Zr-89]Zr-DFO-MSLN-mAb had higher tumor uptake compared to [Zr-89]Zr-3,2-HOPO-MSLN-mAb, while the latter had consistently higher femur uptake. Imaging results were consistent with biodistribution studies.

Towards the stable chelation of radium for biomedical applications with an 18-membered macrocyclic ligand

Summary: Targeted alpha therapy with [Ra-223]RaCl2 has been effective in treating castrate-resistant prostate cancer bone metastases, but efforts to direct [Ra-223]Ra2+ to non-osseous disease sites have had limited success. This study demonstrates the stability and efficiency of the radiocomplex [Ra-223][Ra(macropa)] in directing Ra-223 to non-bone metastases. Conjugation of macropa with beta-alanine and DUPA shows potential for delivering Ra-223 to metastases outside of the bone, expanding the therapeutic utility of this radiometal.
