Multifunctional metal-organic framework (MOF)-based nanoplatforms for cancer therapy: from single to combination therapy

Summary: Cancer remains a severe threat to human health. The marriage of nanomaterials and therapeutic methods, especially metal-organic frameworks (MOFs), provides a practical possibility to improve the deficiencies in cancer therapy. MOFs have been widely applied in cancer therapy to overcome the drawbacks of other therapeutic methods through their controllable morphologies, tailorable diameters, diverse compositions, tunable porosities, high specific surface areas, facile functionalization, and good biocompatibility.

Photothermal Enhanced and Tumor Microenvironment Responsive Nanozyme for Amplified Cascade Enzyme Catalytic Therapy

Summary: This study presents a biodegradable nanocatalyst, PEGylated CuxMnySz (PCMS), that can promote cascade catalytic reactions in tumor microenvironment (TME) with excellent enzyme-like activities and biosafety for tumor therapy. PCMS catalyzes the cascade conversion of H2O2 to oxygen (O-2) and superoxide radical (center dot O-2(-)) in the TME, and exhibits strong photothermal effect under NIR-II 1064 nm laser irradiation, enhancing the chemodynamic therapy (CDT) efficacy. It also benefits magnetic resonance imaging, photoacoustic imaging and infrared thermal imaging to track the fate of PCMS in vivo.

Chirality-Dependent Reprogramming of Macrophages by Chiral Nanozymes

Summary: By designing and synthesizing chiral MoS2/CoS2 nanozymes, their chirality-specific interactions with biological systems efficiently modulate the polarization of tumor-suppressing (M1) tumor-associated macrophages (TAMs), reversing tumor immunosuppression. D (right-handed) nanozymes show longer circulating half-life and higher tumor accumulation, efficiently repolarizing TAMs into M1 phenotype. On the contrary, L (left-handed) nanozymes exhibit limited M1 polarization performance due to chirality-driven homologous adhesion.

Dimensionality Engineering of Single-Atom Nanozyme for Efficient Peroxidase-Mimicking

Summary: This study reports a dimensionality engineering strategy to convert conventional 2D Fe-N-4 centers into 3D structures by integrating oxidized sulfur functionalities onto the carbon plane. The results suggest that oxidized sulfur functionalities can serve as binding sites for assisting substrate orientation and facilitating the desorption of H2O, resulting in an outstanding specific activity of up to 119.77 U mg(-1), which is 6.8 times higher than that of conventional FeN4C SAzymes.

Hierarchical MOF-on-MOF Architecture for pH/GSH-Controlled Drug Delivery and Fe-Based Chemodynamic Therapy

Additionally, the nanomaterial exhibited peroxidase-like catalytic activity and significant inhibitory effects on cancer cells.

Preparation of nanoscale cationic metal-organic framework Nano Mn(iii)-TP for theranostics based on valence changes

However, a Mn-based nano-cation metal-organic framework has shown promise in addressing this challenge by utilizing the over-expression of glutathione in the tumor environment to achieve an outstanding tumor imaging performance and Fenton-like effect. When loaded with the anticancer drug methotrexate disodium, the nanoplatform specifically releases the drug in response to different pH and temperature conditions, leading to an enhanced tumor suppressive effect. This multimodal therapeutic agent holds potential for clinical tumor treatment.

MOF-derived yolk-shell Co@ZnO/Ni@NC nanocage: Structure control and electromagnetic wave absorption performance
