Allele-aware chromosome-level genome assembly of Artemisia annua reveals the correlation between ADS expansion and artemisinin yield

Baosheng Liao et al.

Summary: This study investigates the biosynthesis and regulation of artemisinin through chromosome-level genome mapping. The research discovers a correlation between artemisinin content and the copy number of amorpha-4,11-diene synthase genes, providing new insights into the production of this anti-malarial drug.

Differential transcriptome analysis of genes associated with the rhizome growth and sesquiterpene biosynthesis in Atractylodes macrocephala

Qingyan Ruan et al.

Summary: This study conducted metabolic analysis and transcriptome sequencing of different tissues of Atractylodes macrocephala, identifying key transcription factors involved in regulating rhizome growth and sesquiterpene biosynthesis. EF-1α was found to be the best reference gene for tissue expression pattern analysis. The extensive transcriptome data provided a valuable resource for further research on molecular mechanisms of rhizome growth and sesquiterpene biosynthesis.

The transcription factor OpWRKY2 positively regulates the biosynthesis of the anticancer drug camptothecin in Ophiorrhiza pumila

Xiaolong Hao et al.

Summary: The limited bioavailability of plant-derived natural products with anticancer activity poses challenges to the pharmaceutical industry. Recent study reveals that OpWRKY2 acts as a direct positive regulator of camptothecin biosynthesis, which can increase camptothecin levels by more than three-fold. This finding provides a feasible strategy for the over-accumulation of camptothecin in a biotechnologically amenable system.

AaWRKY9 contributes to light- and jasmonate-mediated to regulate the biosynthesis of artemisinin in Artemisia annua

Xueqing Fu et al.

Summary: AaWRKY9 positively regulates artemisinin biosynthesis by directly binding to the promoters of AaDBR2 and AaGSW1. In the light pathway, the key regulator AaHY5 plays a critical role in activating the expression of AaWRKY9. Additionally, AaWRKY9 interacts with AaJAZ9 in the JA signaling pathway, and the transcriptional activation activity of AaWRKY9 is increased in the presence of methyl jasmonate.

An R2R3-MYB Transcription Factor Positively Regulates the Glandular Secretory Trichome Initiation in Artemisia annua L.

Wei Qin et al.

Summary: The AaMYB17 gene has been identified as a positive regulator of GST initiation in Artemisia annua leaves, leading to increased artemisinin content. Overexpression or RNA interference of AaMYB17 does not affect plant growth or trichome morphology.

An HD-ZIP-MYB complex regulates glandular secretory trichome initiation in Artemisia annua

Lihui Xie et al.

Summary: The study identified key transcription factors, AaMYB16 and AaMYB5, that interact with AaHD1 to regulate glandular secretory trichomes (GSTs) initiation in plants. AaMYB16 was found to be a positive regulator while AaMYB5 had an inhibitory effect on GST initiation. The competitive interaction between AaMYB16 and AaMYB5 modulates the binding activity of AaHD1 promoter, providing new insights into the regulatory network controlling GST formation.

AaMYB15, an R2R3-MYB TF in Artemisia annua, acts as a negative regulator of artemisinin biosynthesis

Zhangkuanyu Wu et al.

Summary: Artemisinin is a secondary metabolite extracted from Artemisia annua, with extensive economical values as an effective antimalarial component. AaMYB15 is identified as the first negative regulator of artemisinin biosynthesis in A. annua, inhibiting the expression levels of key enzyme genes and decreasing artemisinin contents. AaMYB15 directly binds to the promoter of AaORA, a positive regulator, to repress its transcriptional activity and negatively regulate artemisinin biosynthesis.
