Bi-allelic variants in ASTL cause abnormal fertilization or oocyte maturation defects

Yang Zeng et al.

Summary: Fertilization is a critical process in development, and the inhibition of additional sperm binding after fertilization is achieved through mechanisms acting at the zona pellucida and egg plasma membrane. This study identifies bi-allelic variants in the ASTL gene, which are associated with fertilization problems in humans. The identified variants in ASTL significantly decrease the protein levels or affect the enzymatic activity involved in preventing polyspermy. Knock-in mice carrying these variants show subfertility and reduced embryo developmental potential. These findings highlight the role of ASTL in female infertility and provide a new genetic marker for diagnosing fertilization problems.

Experience analysing over 190,000 embryo trophectoderm biopsies using a novel FAST-SeqS preimplantation genetic testing assay

Lauren Walters-Sen et al.

Summary: This study validated FAST-SeqS as a reliable PGT-A method and demonstrated its ability to detect additional information related to ploidy using SNP analysis. The results showed high sensitivity and specificity of FAST-SeqS in detecting whole-chromosome aneuploidies and identified various chromosomal abnormalities in segmental aneuploidies.

Human 1PN and 3PN zygotes recapitulate all morphokinetic events of normal fertilization but reveal novel developmental errors

Kenji Ezoe et al.

Summary: This study investigates the morphokinetic changes during abnormal fertilization using time-lapse technology. It finds that abnormal fertilization recapitulates the overall choreography of normal fertilization but also reveals novel morphokinetic phenomena. The findings raise scientifically and clinically relevant questions about the physiology and pathology of early human development.

Obstetric and neonatal outcomes after the transfer of vitrified-warmed blastocysts developing from nonpronuclear and monopronuclear zygotes: a retrospective cohort study

Ming Li et al.

Summary: The study compared obstetric and neonatal outcomes after transferring vitrified-warmed single blastocysts developing from nonpronuclear (OPN) and monopronuclear (1PN) zygotes with those developing from two-pronuclear (2PN) zygotes. The results showed that while OPN blastocyst transfers resulted in higher birthweights and a greater proportion of very large for gestational age newborns compared to 2PN blastocyst transfers, 1PN blastocyst transfers had a higher abortion rate and lower live birth rate. The study suggests a need for caution when considering 1PN and OPN embryo transfers.
