Multiple interactions of the dynein-2 complex with the IFT-B complex are required for effective intraflagellar transport

Shunya Hiyamizu et al.

Summary: The dynein-2 complex is transported anterogradely within cilia to drive retrograde trafficking of the intraflagellar transport (IFT) machinery containing IFT-A and IFT-B complexes. There are multiple interactions between the dynein-2 and IFT-B subunits, including WDR60 and the DYNC2H1-DYNC2LI1 dimer from dynein-2, and IFT54 and IFT57 from IFT-B. These interactions play a crucial role in the connection between dynein-2 and IFT-B.

The molecular structure of IFT-A and IFT-B in anterograde intraflagellar transport trains

Samuel E. Lacey et al.

Summary: In this study, a molecular model of the anterograde train in cilia assembly was generated using cryo-electron tomography and AlphaFold2 protein structure predictions. The conformations of IFT-A and IFT-B were found to depend on lateral interactions with neighboring repeats, indicating the importance of polymerization for complex stability. Furthermore, IFT-B was shown to extend flexible tethers to maintain a connection with IFT-A under mechanical stresses. These findings provide insights into the fundamental processes of cilia assembly.

Disease-associated mutations in WDR34 lead to diverse impacts on the assembly and function of dynein-2

Caroline Shak et al.

Summary: The primary cilium is a sensory organelle that receives signals from the external environment and transports them into the cell. Mutations in proteins required for transport in the primary cilium result in ciliopathies, which lead to the malformation of organs and skeletal dysplasias. WDR34, a dynein-2 intermediate chain, is necessary for the maintenance of cilia function.

IFT74 variants cause skeletal ciliopathy and motile cilia defects in mice and humans

Zeineb Bakey et al.

Summary: This study explores the function of IFT74 through the investigation of mouse and human alleles. The results show that IFT74 plays an important role in maintaining particle structure and transporting cargos into the cilium. Mutations in IFT74 affect the function of motile cilia but have minimal impact on non-motile cilia.

Summary: It has been discovered that pathogenic DYNC2LI1 deletion variants are compromised in their ability to interact with DYNC2H1 and WDR60, and cause ciliary defects when coexpressed with a missense variant that mimics the situation of compound heterozygous ciliopathy cells.

IFT-A structure reveals carriages for membrane protein transport into cilia

Sophie J. Hesketh et al.

Summary: In this study, researchers successfully reconstituted the complete human IFT-A complex and determined its structure using cryo-EM. They discovered how IFT-A interacts with IFT-B and uses specific domains to create carriages for protein transport in the IFT train. The results also highlight the essential role of IFT-A and TULP adaptor proteins in cilia localization and regulation of protein transport speed by the key kinase ICK.
