Targeting the NRF2/KEAP1 pathway in cervical and endometrial cancers

Summary: Cervical and endometrial cancers are highly dangerous gynecological malignancies with high fatality and recurrence rates. The NRF2/KEAP1 pathway is important in protecting cells against oxidative damage. However, its activation in cancer cells leads to chemoresistance. This review discusses the role of the NRF2/KEAP1 pathway in cervical and endometrial cancers, its components and downstream genes, and strategies to modulate this pathway for chemotherapy.

Research on apoptotic mechanism and related pathways involved in postmortem grass carp (Ctenopharyngodon idellus) muscle

Summary: This study investigated the apoptotic mechanism of postmortem fish during low-temperature storage. The activation of caspases, release of cyt-c, changes in Bcl-2 and Bax protein levels, and transcriptional regulation of various molecules were observed. Both extrinsic and intrinsic apoptotic pathways were involved in the activation of apoptosis, and the translocation of AIF indicated the involvement of a caspase-independent pathway.

The impact of oxidative stress-induced mitochondrial dysfunction on diabetic microvascular complications

Summary: Diabetes mellitus (DM) is a metabolic disease characterized by chronic hyperglycemia, with insulin deficiency or resistance as the main cause, leading to damage in various target organs. Oxidative stress, resulting from an imbalance between free radicals production and antioxidant scavenging, is one of the main mechanisms linking DM and its complications. This paper reviews the dysfunction of mitochondria induced by high glucose-induced oxidative stress in the DM model and proposes a new treatment plan based on mitochondrial dysfunction.

JI017 Induces Cell Autophagy and Apoptosis via Elevated Levels of Reactive Oxygen Species in Human Lung Cancer Cells

Summary: Lung cancer is a common and deadly malignancy. This study tested the effectiveness of a natural drug, JI017, in lung cancer cells. The results showed that JI017 inhibited cell proliferation, induced apoptosis, regulated apoptotic molecules, and inhibited colony formation. JI017 also increased intracellular ROS generation and downregulated PI3K, AKT, and mTOR expression. In addition, JI017 promoted apoptosis through ROS-induced autophagy. In a mouse model, JI017 treatment reduced tumor size and increased markers of apoptosis and autophagy. Targeting JI017 and autophagy signaling could be a potential strategy for lung cancer treatment.

Through the combined evolution of medicinal chemistry and molecular biology, innovative inducers of apoptosis and inhibitors of anti-apoptotic regulators have been discovered. Many of these agents are now being tested in early-phase trials in cancer patients. We believe that apoptosis targeting holds promise as a relevant strategy in cancer therapeutics.

Atractylenolide III induces apoptosis by regulating the Bax/Bcl-2 signaling pathway in human colorectal cancer HCT-116 Cells in vitro and in vivo

Summary: Atractylenolide III, a component of atractylodes, has been found to inhibit cell growth and induce apoptosis in colon cancer cells. Mechanistic studies revealed that it regulates the Bax/Bcl-2 apoptotic pathway. In vivo experiments on nude mice showed that atractylenolide III significantly inhibits tumor growth. These findings provide a scientific basis for understanding the anticancer effect of atractylenolide III and suggest its potential as a promising drug for colorectal cancer treatment.

Apoptin treatment also leads to a significant increase in cellular ROS levels, which are involved in apoptosis and autophagy of liver cancer cells. The increase in ROS levels may be a key factor causing apoptosis and autophagy.

Studies show that mitochondrial infusion can effectively reduce glycolysis, enhance oxidative phosphorylation, reduce proliferation, and promote apoptosis as anti-tumor therapies.
