Synthesis and characterisation of fluorescent substrates for eukaryotic protein N-glycosylation

Mario M. de Capitani et al.

Summary: We report the synthesis of two fluorescently labelled analogues of C25 dolichol (Dol25), which can be accepted by eukaryotic protein N-glycosylation enzymes. The fluorescent labels facilitate handling and purification of the lipid-linked glycosyl donors and acceptors, and should facilitate further biochemical studies of protein glycosylation enzymes.

Molecular basis for glycan recognition and reaction priming of eukaryotic oligosaccharyltransferase

Ana S. Ramirez et al.

Summary: Oligosaccharyltransferase (OST), the central enzyme in N-glycosylation, has been studied using cryo-electron microscopy to reveal its molecular mechanism of substrate recognition and catalysis. The study also discovered that OST can efficiently process closely spaced N-glycosylation sites through alternate priming.

Substrate specificities and reaction kinetics of the yeast oligosaccharyltransferase isoforms

Jillianne Eyring et al.

Summary: The study investigated the biochemical properties of two different OST complexes from Saccharomyces cerevisiae, revealing significant differences in turnover rates for peptide and LLO substrates. This provides a basis for further functional analysis of the Ost3p and Ost6p subunits.
