Phenotypic plasticity and reduced tissue retention of exhausted tumor-infiltrating T cells following neoadjuvant immunotherapy in head and neck cancer

Summary: Neoadjuvant immunotherapies (NITs) show clinical benefits in various cancers and understanding the molecular mechanisms behind the response to NIT can improve treatment strategies. This study reveals that exhausted tumor-infiltrating CD8+ T cells respond to concurrent TGF-0 and PD-L1 blockade, leading to increased circulating Tex cells and reduced CD103 expression. TGF-0 plays a role in tissue retention and impaired systemic immunity, and transcriptional changes in T cell receptor signaling and glutamine metabolism affect the response to NIT.
