Genomic characterization of coexisting anatoxin-producing and non-toxigenic Microcoleus subspecies in benthic mats from the Wolastoq, New Brunswick, Canada

Summary: The presence of toxigenic benthic cyanobacteria in riverine ecosystems is a global concern. In 2018, the death of three dogs in Canada was linked to anatoxin exposure from benthic microbial mats. DNA sequencing of cyanobacterial isolates from the mat samples revealed that some produced anatoxins while others did not. Genomic comparisons showed that toxigenic and non-toxigenic Microcoleus subspecies coexisted with differences in the presence/absence of the anatoxin-a biosynthesis cluster.

Recovering metagenome-assembled genomes from shotgun metagenomic sequencing data: Methods, applications, challenges, and opportunities

Summary: Reference genomes are crucial for analyzing the metabolic and functional potentials of microbiomes. However, limited microbial genome resources are available due to the difficulty of culturing microorganisms. Genome binning is a culture-independent method that can recover a large number of microbial genomes from short-read high throughput shotgun metagenomic sequencing data. In this review, commonly used methods for reconstructing metagenome-assembled genomes (MAGs) are summarized to assist researchers in selecting appropriate software programs for these analyses. Additionally, the application prospects, challenges, and opportunities for recovering MAGs from metagenomic sequencing data are discussed.

Genome sequence of freshwater nontoxigenic Limnoraphis associated with microcystin-producing blooms.

Summary: A sample from a cyanobacterial bloom in a freshwater reservoir in eastern Oregon was used to obtain the complete genome of Limnoraphis sp. WC205. This cyanobacterium coexisted with other cyanobacteria and was associated with microcystin production, although it does not possess the genes for producing cyanotoxins. The microcystin producer in the reservoir was identified as Microcystis through DNA sequencing.
