Loss of IGFBP2 mediates alveolar type 2 cell senescence and promotes lung fibrosis

Chiahsuan Chin et al.

Summary: Accumulation of senescent cells contributes to age-related diseases including idiopathic pulmonary fibrosis (IPF). Intranasal delivery of recombinant IGFBP2 protects aged mice from weight loss and demonstrated antifibrotic effects after bleomycin lung injury. Expression of IGFBP2 is suppressed in AEC2 cells isolated from fibrotic lung regions of patients with IPF and/or pulmonary hypertension compared with patients with hypersensitivity pneumonitis and/or chronic obstructive pulmonary disease.

Calming the cytokine storm of COVID-19 through inhibition of JAK2/STAT3 signaling

Bharath Kumar Gajjela et al.

Summary: The outbreak of SARS-CoV-2 has caused the COVID-19 pandemic, infecting over 178 million people worldwide. Despite new vaccines, achieving global herd immunity remains a challenge. Severe COVID-19 patients suffer from respiratory failure due to acute respiratory distress syndrome, a leading cause of mortality. Lung hyper-inflammation caused by virus-induced tissue damage and uncontrolled cytokine release is a major factor. FDA-approved JAK inhibitors are being evaluated and repurposed to treat severe COVID-19 by reducing SARS-CoV-2 infection.

Identification and Validation of Aging-Related Genes in Idiopathic Pulmonary Fibrosis

Jie He et al.

Summary: In this study, 16 potential aging-related genes associated with idiopathic pulmonary fibrosis (IPF) were identified and validated using bioinformatics analysis and clinical sample validation. Among these genes, IGF1, RET, and IGFBP2 were found to play a role in the occurrence and prognosis of IPF. The findings may contribute to the understanding of IPF pathogenesis.

Updates on community acquired pneumonia management in the ICU

Girish B. Nair et al.

Summary: Despite the global pandemic related to SARS-CoV-2, Streptococcus pneumoniae infection remains the most common cause of severe community acquired pneumonia. Timely diagnostics and adequate antibiotic coverage are crucial for reducing mortality, while novel antibiotics offer additional therapy options for patients not responding to standard treatment.

IGFBP2 promotes tumor progression by inducing alternative polarization of macrophages in pancreatic ductal adenocarcinoma through the STAT3 pathway

Longhao Sun et al.

Summary: This study revealed the role of IGFBP2 in TAM polarization, where IGFBP2 promotes the accumulation of M2-like TAMs and plays a pro-tumor role in PDAC growth. Mechanistically, IGFBP2 activates STAT3 to increase IL-10 secretion, driving TAM polarization towards an M2 phenotype and contributing to the immunosuppressive microenvironment in PDAC.

Macrophages in Lung Injury, Repair, and Fibrosis

Peiyong Cheng et al.

Summary: Fibrosis progression in the lung can lead to impaired gas exchange, respiratory failure, or even death. Macrophages play a crucial role in response to lung injury, repair, and fibrosis, with their activation and polarization being key factors in the process. Targeting macrophages may offer potential benefits in alleviating pulmonary fibrosis.
