Extracellular ATP: A powerful inflammatory mediator in the central nervous system

Summary: Nucleotides, especially extracellular ATP (eATP), play a crucial role in inter-species extracellular signaling. Different organisms have evolved sensors with varying affinity and selectivity for eATP, resulting in a highly flexible signaling system. In mammalian evolution, the control of intracellular ATP and eATP homeostasis is closely linked to that of Ca2+. Specific plasma membrane receptors, known as P2 receptors (P2Rs), sense eATP with various degrees of affinity and selectivity, adding to the plasticity of the system. The central nervous system (CNS) is a privileged site for purinergic signaling, with all brain cell types expressing P2Rs. Evidence suggests that eATP not only participates in synaptic transmission but also plays a crucial homeostatic role in brain cell responses. Modulating eATP concentration in the CNS may hold promise for the treatment of neuroinflammation.

An insight into new glycotherapeutics in glial inflammation: Understanding the role of glycosylation in mitochondrial function and acute to the chronic phases of inflammation

Summary: This study investigated the glycosylation modulation in a mixed glial culture model and identified considerable differences in glycosylation from acute to chronic phases. Clustering analysis revealed dense clusters for control and LPS treated groups, while the cytokine combination group was scattered into different clusters. Lectin immunostaining confirmed the presence of various glycans in astrocytes and microglia, including GalNAc, GlcNAc, mannose, fucose, and sialic acid-binding residues. Inhibition of sialyltransferase suppressed the upregulation of sialic acid expression, resulting in modulation of mitochondrial functions.

Choroid plexus-selective inactivation of adenosine A2A receptors protects against T cell infiltration and experimental autoimmune encephalomyelitis

Summary: A(2A)R is upregulated in the choroid plexus (CP), enhancing its gateway activity in experimental autoimmune encephalomyelitis (EAE) mice. Treatment with the A(2A)R antagonist KW6002 reduces T cell trafficking across the CP and attenuates EAE pathology. Focal knock-down of CP-A(2A)R inhibits the infiltration of Th17(+) cells across the CP through the CCR6-CCL20 axis, leading to protection against EAE pathology. Activation of A(2A)R in the cultured CP epithelium increases CP permeability and facilitates lymphocyte migration.
