UniProt: the universal protein knowledgebase in 2021

Alex Bateman et al.

Summary: The UniProt Knowledgebase aims to provide users with a comprehensive, high-quality set of protein sequences annotated with functional information. Updates over the past two years have increased the number of sequences to approximately 190 million, with new methods to assess proteome completeness and quality. UniProtKB has responded to the COVID-19 pandemic by expertly curating relevant entries and making them rapidly available through a dedicated portal.

Nonframeshifting indel variations in polyalanine repeat of HOXD13 gene underlies hereditary limb malformation for two Chinese families

Bailing Zu et al.

Summary: This study identified two nonframeshifting mutations of HOXD13 polyalanine repeat location in two Chinese families with postaxial polydactyly or postaxial syndactyly through whole exome sequencing. These mutations led to truncation or expansion of Poly-Ala repeats of HOXD13 proteins and resulted in conformational changes according to three-dimensional structural analysis. The findings provide new insights into genetic counseling and clinical management.

A Novel Missense Variant of HOXD13 Caused Atypical Synpolydactyly by Impairing the Downstream Gene Expression and Literature Review for Genotype-Phenotype Correlations

Ruiji Guo et al.

Summary: Synpolydactyly (SPD) is a hereditary congenital limb malformation caused by mutations in the HOXD13 gene. Research has shown that different mutations in HOXD13 are associated with the severity and symptoms of SPD, providing insights for clinical diagnosis and genetic counseling.

GLI3 variants causing isolated polysyndactyly are not restricted to the protein's C-terminal third

Henrike Lisa Sczakiel et al.

Summary: Loss of function variants of GLI3 are associated with different types of polysyndactyly, with GCPS and IPD showing varying severity levels of the same disorder. Variants affecting GLI3 have been found in patients with clinical findings suggestive of a GLI3-associated syndrome, confirming the association of PHS with variants affecting the central third of GLI3 protein. The study suggests a binary division of GLI3-associated disorders into PHS or GCPS/polysyndactyly.

Genome sequencing in families with congenital limb malformations

Jonas Elsner et al.

Summary: By conducting genome sequencing on 69 undiagnosed patients with congenital limb malformations, this study identified multiple potentially pathogenic genetic variants, including both known disease genes and novel disease genes. Additionally, complex structural variants were found, but no causative noncoding variants were identified.

Highly accurate protein structure prediction with AlphaFold

John Jumper et al.

Summary: Proteins are essential for life, and accurate prediction of their structures is a crucial research problem. Current experimental methods are time-consuming, highlighting the need for accurate computational approaches to address the gap in structural coverage. Despite recent progress, existing methods fall short of atomic accuracy in protein structure prediction.
