As a Novel Tumor Suppressor, LHPP Promotes Apoptosis by Inhibiting the PI3K/AKT Signaling Pathway in Oral Squamous Cell Carcinoma

Shanshan Liu et al.

Summary: Oral squamous cell carcinoma (OSCC) is a common malignant tumor of the head and neck with a poor prognosis and unsatisfactory treatment. This study found that the protein histidine phosphatase LHPP plays an important role in the development of OSCC. LHPP inhibits the growth and migration of OSCC by decreasing the activity of the PI3K/AKT signaling pathway and promoting apoptosis.

Agrin Promotes Non-Small Cell Lung Cancer Progression and Stimulates Regulatory T Cells via Increasing IL-6 Secretion Through PI3K/AKT Pathway

Linzhi Han et al.

Summary: The study examined the effects and mechanisms of Agrin in non-small cell lung cancer (NSCLC) and the tumor immune microenvironment. It was found that Agrin promotes tumor cell growth and infiltration of regulatory T cells (Tregs) by increasing IL-6 expression and secretion through the PI3K/AKT pathway. Knockdown of Agrin can suppress the proliferation and metastasis of NSCLC cells.

Shuang Wu et al.

Summary: This study identifies ZC3H13 as a potential tumor suppressor gene in hepatocellular carcinoma (HCC) and reveals the miR-362-3p/miR-425-5p-ZC3H13 axis as a regulatory pathway. The expression of ZC3H13 is closely associated with the prognosis and tumor immune infiltration in HCC.

Sho Tsukiyama et al.

Summary: In this study, a novel approach called BERT6mA was proposed for detecting the 6mA modification in DNA, and its performance was evaluated and compared. Through pretraining and fine-tuning, BERT6mA showed high performance in prediction and achieved good results even in species with small sample sizes. Furthermore, the study analyzed the process of feature extraction by the BERT6mA model.

ebv-circRPMS1 promotes the progression of EBV-associated gastric carcinoma via Sam68-dependent activation of METTL3

Jing-yue Zhang et al.

Summary: This study found that highly expressed ebv-circRPMS1 in EBVaGC can promote the proliferation, migration, and invasion of cancer cells, and inhibit apoptosis, while METTL3 was upregulated. Mechanistically, ebv-circRPMS1 can bind to Sam68 to facilitate its interaction with the METTL3 promotor, resulting in the transactivation of METTL3.

Therapeutic targeting m6A-guided miR-146a-5p signaling contributes to the melittin-induced selective suppression of bladder cancer

Rucheng Yan et al.

Summary: Abnormal RNA methylation and dysregulation of miRNA are common in bladder cancer, and melittin has shown potential as a drug candidate for intravesical chemotherapy against bladder cancer. METTL3 has been identified as a key regulator in controlling bladder cancer cells' sensitivity to melittin-induced apoptosis, while miR-146a-5p suppression contributes to the selective antitumor effect of melittin.

RNA demethylase ALKBH5 inhibits TGF-β-induced EMT by regulating TGF-β/SMAD signaling in non-small cell lung cancer

Zelong Sun et al.

Summary: ALKBH5 plays a key role in regulating TGF-beta/SMAD signaling and controlling TGF-beta-induced EMT in NSCLCs. ALKBH5 expression is significantly reduced in metastatic NSCLC and its overexpression inhibits TGF-beta-induced EMT and invasion of NSCLC cells. ALKBH5 affects the expression and mRNA stability of TGF beta R2, SMAD3, and SMAD6 by erasing m(6)A modification in NSCLC cells, and interacts with m(6)A-binding proteins to further regulate TGF-beta signaling.

Summary: Accumulating evidence suggests that m6A modifications play crucial roles in colorectal cancer (CRC) by regulating genes involved in glucose metabolism, RNA metabolism, and cancer stem cells. Dysregulated m6A peaks in CRC samples are associated with tumour progression and patient survival, highlighting the potential of modulating m6A modifications as a strategy for predicting survival and interfering with tumour progression in cancer patients.

Improving function of cytotoxic T-lymphocytes by transforming growth factor-β inhibitor in oral squamous cell carcinoma

Summary: Immunotherapy with immune-checkpoint therapy for OSCCs is limited by the immunosuppressive effects of TGF-beta, which inhibits CTL function; TGF-beta inhibitor effectively restores CTL function and is primarily expressed at the tumor invasive front; TGFB1 mRNA expression is significantly negatively correlated with the CD8(+) T-cell/Treg ratio and Ki-67 expression in CD8(+) T-cells, indicating TGF-beta suppresses CTL function in situ.
