Coumarin-pyrazoline Hybrids as Selective Inhibitors of the Tumor-associated Carbonic Anhydrase IX and XII

Summary: This study synthesized a series of coumarin derivatives incorporating pyrazole-1-carboxamide moiety and evaluated their inhibitory activity against carbonic anhydrase. Antimicrobial and Antibiofilm Activities of Carvacrol, Amoxicillin and Salicylhydroxamic Acid Alone and in Combination vs. Helicobacter pylori: Towards a New Multi-Targeted Therapy

Summary: The World Health Organization has identified Helicobacter pylori as a high-priority pathogen requiring updated antibacterial treatments. This study investigated the potential of combining a CA inhibitor, carvacrol (CAR), amoxicillin (AMX), and a urease inhibitor (SHA) to develop a multiple-targeted anti-H. pylori therapy. The combinations of these compounds showed strong inhibition of H. pylori growth and biofilm formation, with CAR-AMX and CAR-SHA combinations demonstrating additive effects and AMX-SHA combination showing indifferent effects.

The software allows rapid generation of libraries of bioactive ligands, building blocks, or commercial compounds by manual creation or importing from public databases. The databases can be enriched with various data and filtered based on molecular structures or properties, making the desired molecules easily accessible. Additionally, the software can predict molecular properties and potential toxicological effects. MolBook UNIPI is an invaluable tool for medicinal chemists, even for inexperienced users without cheminformatics knowledge or programming skills. Click ferrocenyl-erlotinib conjugates active against erlotinib-resistant non-small cell lung cancer cells in vitro

Summary: A library of 1,4- and 1,5-triazole ferrocenyl derivatives of erlotinib were synthesized and their anticancer activity was studied against erlotinib-sensitive and erlotinib-resistant lung cancer cells. Among the compounds investigated, two isomers showed superior activity against erlotinib-resistant cells, with a mechanism of action different from erlotinib.

Focused library of phenyl-fused macrocyclic amidinoureas as antifungal agents

Summary: The outcomes of hospitalized patients for invasive fungal infections are worsening due to the increase in antimicrobial-resistant phenotypes and the spread of the global pandemic of COVID-19. Among these infections, candidiases are particularly concerning as the available drugs are extremely limited. A new class of macrocyclic amidinoureas with a guanidino tail has shown promise as antifungal agents. In this study, a focused library of seven derivatives of macrocyclic amidinoureas, with a second phenyl ring fused with the core, was designed and synthesized. Some compounds showed interesting antifungal activity against a variety of Candida spp. and C. neoformans. The compounds had a low passive diffusion, suggesting a membrane-based mechanism of action or the involvement of active transport systems. The compounds were also found to be non-toxic at high concentrations.

Discovery of a Series of 1,2,3-Triazole-Containing Erlotinib Derivatives With Potent Anti-Tumor Activities Against Non-Small Cell Lung Cancer

Summary: In this study, novel Erlotinib derivatives containing 1,2,3-triazole rings were synthesized by linking Erlotinib to azide compounds. These compounds showed strong antiproliferation activity against various NSCLC cells, with two of them being more effective than Erlotinib in most cell lines. They induced apoptosis and cell cycle arrest in PC-9 and H460 cells and exhibited inhibitory activities on EGFR phosphorylation. These Erlotinib-1,2,3-triazole compounds are potential candidates for NSCLC treatment and warrant further investigation.
